| The main cause of deep vein thrombosis(DVT)is abnormal coagulation of venous blood in the deep venous cavity and attachment to the vessel wall,resulting in obstruction of the vein and obstruction of reflux.When the thrombus falls off under the action of fibrinolytic enzyme,it may cause pulmonary embolism.(PE),if the condition is serious,it will be life-threatening,and most of the emboli(embolus)that cause pulmonary embolism are from the deep veins of the lower extremities.Therefore,in-depth study of the mechanism of deep vein thrombosis of the lower extremities will be important for clinical screening,diagnosis and treatment.significance.Omics technology is currently an important tool for systems biology research.Through the overall analysis of biological systems,it is possible to study the interaction mechanism and internal connections between biological systems and diseases,and to find important disease-related markers and drug treatments.Target.Research purpose:There are no related literature reports on the differential function of the intestinal flora and metabolic regulation network in patients with deep vein thrombosis of the lower extremities,which hinders the complete understanding of the related etiology and biomechanics of the pathogenesis of deep vein thrombosis of the lower extremities.The omics technique preliminarily explored the formation mechanism of deep vein thrombosis of lower limbs.Research method:In this study,31 patients with deep vein thrombosis of lower limbs and 20 healthy volunteers were recruited in the Second Department of Vascular Surgery Department of the First Hospital of Jilin University.All subjects are between 18 and 80 years old,have no history of smoking,have no diabetes,other blood diseases,intestinal diseases,metabolic diseases and other diseases affecting the intestinal flora and have not received any of them for more than 2 hours recently Surgery,no hormone therapy,no liver and kidney dysfunction and other contraindications.All stool samples and some blood samples of patients and healthy volunteers were collected through microbiology and metabolomics.The method found several metabolic pathways that may affect the occurrence of deep vein thrombosis while identifying the differences in the flora,and initially predicted the possible pathogenesis of deep vein thrombosis.Conclusion:The results of microbiology analysis showed that deep vein thrombosis of lower extremities is closely related to intestinal flora disorder.The composition and structure of intestinal flora of patients with deep vein thrombosis of lower extremities are significantly different from those of normal control group.Patients with Bacteroides,Bifidobacteria,The relative abundance of normal flora such as Firmicutes and Actinomyces decreased,and the relative abundance of conditional pathogenic bacteria such as Enterococcus and Enterobacter increased.Patients with deep vein thrombosis of the lower extremities may be due to the increased abundance of gram-negative harmful bacteria such as Enterobacter,and the decreased abundance of normal flora such as bifidobacteria and Bacteroides destroys the integrity of the intestinal mucosal barrier and causes the flora to produce short chains Fatty acid capacity is reduced,thereby affecting the TMA-TMAO metabolic pathway,leading to the formation of deep vein thrombosis of the lower extremities.The results of metabonomics analysis showed that the content of many endogenous metabolites in the serum of the disease group was significantly changed compared with the normal group,and 5 different metabolites and 6 related metabolic pathways were found in blood samples.Among them,the metabolites of arachidonic acid metabolism,linoleic acid metabolism and amino acid pathway may be the marker metabolites of deep vein thrombosis of lower limbs. |
