Background:Lung cancer is a kind of malignant tumor,which is clinically common in bronchi and alveoli,so it is also called bronchoalveolar carcinoma.At present,lung cancer ranks first in the incidence and mortality of malignant tumors in the world.Clinically,lung cancer includes squamous cell carcinoma,adenocarcinoma and large cell lung cancer.Because lung cancer cell proliferation,infiltration and metastasis are a very complex biological behavior,it involves the interaction between multiple genes and multiple pathways.Therefore,revealing the role of lung cancer in the occurrence and development of lung will contribute to the clinical prevention and treatment of lung cancer.The pathogenesis of lung cancer was always studied by our office,and found that TGF-β1 and TGM2 may play an important role in the occurrence and development of lung cancer.Transforming growth factor-β1(TGF-β1)is a member of TGF-βligand that has two-way regulation on tumor cell proliferation,growth,migration,and invasion.In the early stage of tumorigenesis,TGF-β1 is a factor that mainly inhibits tumor growth,and its expression continues to increase,and it has the effect of hindering the proliferation of cancer cells.However,its specific mechanism needs to be further studied.Transglutaminase 2(TGM2),Ca2+-dependent transamidase,is a member of the transglutaminase family.At the same time,its up-regulation is also related to the transcription of HIF-1A.Hypoxia up-regulates the expression of TGM2 through HIF-1a,which can activate the activity of TGM2 in cells.At the same time,increased TGM2 expression in cancer is closely related to drug resistance due to its active nuclear factor-k B(NF-k B)cross-linking and free I-k B polymerization by TGM2.Cisplatin(DDP)is a metal platinum complex,which belongs to cell cycle non-specific anti-tumor drugs.It can damage DNA,inhibit tumor growth,and has a broad antibacterial spectrum.Lobaplatin is a new type of anti-tumor platinum drug developed in recent years.First,the expression of TGF-β1 and TGM2protein was analyzed at the mRNA level.Secondly,the expression of TGF-β1 and TGM2 protein was verified in tissues by histochemical staining.Thirdly,the expression of TGF-β1 and TGM2 in A549 cells treated with cisplatin and lobaplatin.Finally,the expression of TGM2 in A549 cells treated with TGF-β1(ITD-1)inhibitor was analyzed,and then determine the correlation between TGF-β1 and TGM2.Objective:The purpose is to analyze the expression of TGF-β1 and TGM2 in lung cancer cells.Its targeted regulation of TGF-β1 and TGM2 proteins in A549 cells treated with cisplatin and lobaplatin was analyze.Its relationship of TGM2 and TGF-β1 protein was analyzed by regulating TGF-β1 protein,which laid the foundation for the prevention and treatment of lung cancer in the future.Method:1.The online database UCSC was used to collect 1027 lung cancer tissue samples,Analysis of TGF-β1 and TGM2 mRNA expression and prognosis.At the same time,the correlation analysis of the expression information of TGF-β1 and TGM2 genes in lung cancer was carried out by the online database UCSC.2.The positive expression of TGF-β1 and TGM2 protein was analyzed by immunohistochemical staining.3.CCK-8 method was used to analyze the survival rate of cisplatin and lobaplatin on lung cancer A549 cells.4.Flow cytometry was used to analyze the effects of cisplatin and lobaplatin on the cell cycle of lung cancer A549.5.Immunofluorescence technology was used to analyze the expression of TGF-β1 and TGM2 in cells.6.Western blotting was used to analyze the expression of TGF-β1 and TGM2 after treatment of lung cancer A549 cells with cisplatin and lobaplatin.7.Immunofluorescence technology and Western bolt method were used to analyze the expression of TGM2 protein in lung cancer A549 cells treated with TGF-β1inhibitor.Result:1.TGF-β1and TGM2 mRNA are highly expressed in lung cancer tissues.2.The results of immunohistochemistry showed that the expression of TGF-β1and TGM2 in lung cancer tissues was significantly higher than that in adjacent tissues.3.Cisplatin and Lobaplatin can induce apoptosis of lung cancer A549 cells.In addition,they are dose-dependent in A549 cells.4.Cisplatin and Lobaplatin can regulate the cell cycle of lung cancer A549.5.Cisplatin and lobaplatin can reduce the expression of TGF-β1and TGM2 in lung cancer A549 cells.6.The expression of TGM2 decreased in lung cancer A549 cells treated with TGF-β1inhibitor(ITD-1).Conclusions:1.TGF-β1and TGM2 may be closely related when lung cancer occurs.2.TGF-β1 inhibitor may regulate the expression of TGM2 in lung cancer.3.Cisplatin and lobaplatin may play a role in chemotherapy by regulating the expression of TGF-β1 and TGM2.
