| Objective: To understand Alzheimer’s disease(AD)patients,amnestic mild cognitive impairment(aMCI)patients and healthy controls,the peripheral blood Aβ42,p-tau181 protein levels and Helicobacter pylori(HP)infection,to explore the correlation between HP and the levels of Aβ42,p-Tau181 in peripheral blood of AD and aMCI patients.Methods: A total of 90 elderly people who attended the Department of Neurology,Zhongshan Hospital of Dalian University from December 2019 to December 2020 were collected as the research subjects.Among them,there were 27 cases in the AD group,with an average age of(74.70±5.83)years.There were 31 cases in the aMCI group,with an average age of(67.48±6.09)years;32 cases in the control group,with an average age of(64.13±4.27)years.Cognitive assessment uses the Mini Mental State Assessment Scale(MMSE),the Montreal Cognitive Assessment Scale(MOCA),the Clinical Dementia Rating Scale(CDR),the living ability measurement uses the Daily Living Ability Scale(ADL),and the Hamilton Anxiety Scale,Hamilton Depression Scale excludes anxiety and depression;carbon-13-urea breath test,blood HP antibody test for HP test;Aβ42 and p-tau181 protein concentration in peripheral blood are determined by enzyme-linked immunosorbent assay(ELISA)method.Results: 1.With the aggravation of cognitive impairment,serum Aβ42 showed a downward trend.There were statistical differences between the NC group and the AD group,and the NC group and aMCI group(P<0.05);the serum p-tau181 showed a gradual upward trend,but between the groups The difference was not statistically significant(P>0.05);through ROC curve analysis,neither of them had the characteristics of biomarkers for the definite diagnosis of AD.2.The HP infection situation of NC group,aMCI group and AD group was compared.The difference between AD group and NC group,aMCI group and NC group was statistically significant(P<0.05).3.A comparative analysis of the serum Aβ42 and p-tau181 concentrations of HP-positive and HP-negative persons in aMCI and AD groups respectively showed that the serum Aβ42 concentrations of HP-positive persons in aMCI and AD groups were higher than those of HP-negative persons,and there were differences between the groups All were statistically significant(P<0.05);no statistical difference was found in the serum p-tau181 concentration between the groups;4.Analysis of the differences in the serum Aβ42 and p-tau181 concentrations in the aMCI and AD groups for different HP infection levels With the increase of HP infection grade,the serum Aβ42 concentration showed an increasing trend,and the difference between groups was statistically significant(P<0.05);while the serum p-tau181 concentration had no significant trend change,and the difference was not statistically significant(P>0.05).Conclusion: 1.Comparing the serum Aβ42 concentration between AD,aMCI and NC groups,the difference between NC group and AD group,NC group and aMCI group is statistically significant,suggesting that there is a correlation between the peripheral blood Aβ42 level and AD and aMCI.2.Comparison of HP infection in NC group,aMCI group and AD group,the difference between NC group and AD group,NC group and aMCI group is statistically significant,suggesting that HP infection may be correlated with AD and aMCI.3.The serum Aβ42 concentration of patients with HP infection in the aMCI and AD groups was higher than that of those without HP infection,suggesting that HP infection is related to the serum Aβ42 concentration in patients with aMCI and AD.HP infection may affect Aβ42 metabolism in some way,thereby affecting AD. |
