Background Endometriosis is a benign gynecological disease,with the characteristics of malignancy,such as proliferation,invasion and migration,which affects the quality of life of many women.Ovarian endometriosis has a certain malignant potential.Women with histologically proven endometriosis have an increased risk of developing clear cell and endometrioid epithelial ovarian cancer.The pathogenesis of endometriosis is well understood,but remains unclear.Furin is a protease that is involved in a variety of protein activation,known as the main switch of tumor growth and progress,which can be closely related to many cancers and inflammation of many cancers,including neuropeptides,hormones,growth factors and their receptors and adhesion molecules.Objective This experiment was designed to estimate the expression of Furin in the eutopic endometrium of the normal group and ovarian endometriosis group by reverse transcription real-time fluorescent quantitative polymerase chain reaction(RT-qPCR),and to explore the significance of Furin in the development of endometriosis,and provide theoretical evidence for the clinical diagnosis of endometriosis in the ovary.Methods The endometrial tissue of 20 cases of ovarian endometriosis confirmed by pathology was analyzed.The expression of Furin in two groups was detected by RT-qPCR and Western blot,and the potential role of Furin in the development and development of endometriosis was analyzed。Statistics:Combined use of Light Cycler Data Analysis software and MS Excel using 2-ΔΔCTmethod to calculate the ratio difference of the expression level before and after processing.ΔCT(Control group)=c-d,ΔCT(EM group)=a-b,ΔΔCT=ΔCT(EM group)-ΔCT(control group)=(a-b)-(c-d),fold change=2-ΔΔCT=2-((a-b)-(c-d))=N;The statistical analysis was performed by Graph Pad Prism 5,and all statistical analyses were performed by unpaired double-tailed t test.0.05was considered statistically significant;Finally,histograms are done.Results(1)1.Furin mRNA expression:The results of RT-qPCR shows that there are significant differences in mRNA expression in normal intimal tissues and endometriosis tissues,that is,Furin expression level in the former was significantly lower than that in the latter,and the difference was statistically significant(P≤0.01).2.Furin protein expression:Western blotting analysis showed that Furin protein expression was significantly increased in the in-situ intimal tissue of endometriosis patients compared with normal intimal tissue,which verified the difference in Furin expression between the two groups from the protein level.Conclusion Furin expression was increased in the endometriosis group,indicating that Furin is at a low level in normal endometriosis tissues,while Furin overexpression in ovarian endometriosis may activate inflammatory mediators and trigger inflammatory responses.Or Furin hydrolyzes NF-k B and TGF-βthrough its proteolytic enzyme activity,and then induces cell proliferation and inflammatory response.Furin overactivation plays an important role in the occurrence and development of ovarian endometriosis,but its specific mechanism has not been clarified yet,and further experimental proof is needed.
