| Swertia mileensis is traditionally used for protecting liver,and its effective material base is swertiamarin.But due to its structural instability,it is easily decomposed and transformed,so the material base of protecting liver is not clear,and the pathway of transformation in Swertia mileensis is unclear.To study the chemical constituents from Heat-Transformed Swertiamarin.The compounds were isolated and purified by silica gel,reverse phase C18 sili-ca gel,Sephadex LH-20,and semi-prepative HPLC.Their structures were elucidated on the basis of physicochemical properties and spectral data.Twenty-seven compounds including two novel compounds,Sweritranslactone D(1),Sweritranslactone E(2),were isolated and identified.The structures involve iridoids,iridoids dimer,and xanthone.According to the structural correlation between the swertiamarin and heat-transformed swertiamarin,the plausible transformation pathway of chemical constituents from swertiamarin in heating water is proposed in this paper.Moreover,some compounds were evaluated for hepatoprotective activity,and the results showed that liver protective activities of Sweritranslactone D(1)and Sweritranslactone E(2)were better than the swertiamarin,and compared with the positive control group,compound 1 also has strong hepatoprotective activity.In addition,this thesis also reviewed the chemical components of iridoid dimers,and provided references for the research of iridoids. |
