This thesis mainly includes the following two aspects of research content.In the first chapter,the chemical constituents of C.Sinensis was studied.In the second chapter,the chemical constituents and in vitro anti-tumor activity screening of P.ternate were studied.And the research progress of genus Cryptoporus in the past thirty years was summarized in the last part.Objective:To study the chemical compositions of C.Sinensis and P.ternate,and searched for new anti-tumor active substances through in vitro cytotoxic activity.Methods:The compounds were isolated and purified by using various column chromatography methods including silica gel,Rp-18,Sephadex LH-20,MCI and Preparation of thin layer chromatography.1D,2D NMR,HR-ESI-MS,UV and IR spectral analysis were used to identify the structure of the compound.Furthermore using MTS method to study anti-tumor activity of new componds.Results:In the experiment,95% ethanol was used for extraction,followed by extraction with petroleum ether,ethyl acetate and n-butanol.Twenty-three compounds were obtained from Petroleum ether and ethyl acetate part of 95% ethanol extract of fruit body of C.Sinensis,and identified as Cryptoporic acid A methyl ester(1),Cryptoporic acid B(2),Cryptoporic acid B methyl ester(3),Cryptoporic acid D(4),Cryptoporic acid D dimethyl ester(5),Cryptoporic acid E(6),Cryptoporic acid E methyl ester(7),Cryptoporic acid E dimethyl ester(8),Cryptoporic acid G(9),Cryptoporic acid J(10),Cryptoporic acid L(11),Cryptoporic acid N(12),11-hydroxyldrim-8,12-en-14-oic acid(13),Ergosterol(14),5α,8α-peroxyergosta-6,22-diene-3β-ol(15),Ergosta-7,22-dien-3-one(16),trametenolic acid B(17),5α,6α-epoergoster-8(14),22-diene-3β,7α-diol(18),Ergosta-7,22-diene-3β,5α,6β,9α-tetraol(19),Ergosta-7,22-diene-3β,5α,6β-triol(20),Octadecadienoic acid(21),Palmitic acid(22),Glyceryl monopalmitate(23).Nine compounds were obtained from ethyl acetate part of 95% ethanol extract of Tuber of P.ternate,and identified as Pinternatines A(1),Pinternatines B(2),(2S,3S,6S)-2-methyl-6-(9-phenylnonyl)piperidin-3-ol(3),Ephedrine(4),P-hydroxycoumaric acid(5),Chrysophanol(6),Stigmasterol(7),β-sitosterol(8),Linoleic acid-1-monoglyceride(9).Among them,compounds 1 and 2 were two new organic amine compounds,and the anti-tumor active test results show that both compounds 1 and 2 have good cytotoxic activity.Conclusion:The chemical constituents were mainly cryptoporous acid and steroids of C.Sinensis and several cryptoporous acid had been reported with anti-tumor,anti-inflammatory and antioxidant activities.It were Alkaloids containing that isolated from P.ternate,In vitro cytotoxic activity screening found that both compounds 1 and 2have good cytotoxic activity on experimental tumor cell lines,which verifies the rationality of P.ternate anti-tumor effect.
