ObjectiveTo study the difference of IDO1 expression in nasal tissues of patients with chronic rhinosinusitis with nasal polyps and normal inferior turbinate tissues,an d to analyze the expression level of IDO1 in chronic rhinosinusitis with nasal po lyps and its relationship with clinicopathological features,and to explore the sign ificance of this gene as a prognostic index of chronic rhinosinusitis with nasal p olyps.Methods1.The tissues of nasal polyps from 27 patients with chronic sinusitis with na sal polyps(CRSwNP)and functional endoscopic surgery were collected from the first affiliated Hospital of Dali University from January 2020 to June 2020,and the inferior turbinate tissues of 9 patients with simple deviation of nasal septum were collected as the control group.The control group had no history of sinusit is.Lund-Mackey method was used to evaluate the severity of all patients.2.The tissue samples of nasal polyps were collected,and the nasal polyps w ere divided into EOSNP group and NEUNP group according to the degree of eo sinophil infiltration and neutrophil infiltration count by hematoxylin-eosin(HE)st aining.3.The distribution and expression of IDO1 in EOSNP and NEUNP nasal pol yps and normal inferior turbinate tissues were detected by immunohistochemical staining and Western-blotting in 27 cases of nasal polyps(eosinophil 16 cases,n eutrophils 11 cases)and 9 cases of control group.4.The expression of IDO1 mRNA in the three groups was detected by RT-qPCR,and the relative quantitative analysis was carried out byΔCt method.The expression ofΔCt was inversely proportional to that of mRNA,and then the rel ative expression of IDO1 mRNA in EOSNP group,NEUNP group and control g roup was calculated by 2-ΔCtΔCtmethod.5.In this study,SPSS 22.0 software is used to analyze the data.Results1.There was no significant difference in age and sex among EOSNP,NEU NP and control group.There was no significant difference in the Lund-Mackey s core between the EOSNP group and the NEUNP group.The Lund-Mackey score of the control group was lower than that of the EOSNP group and the NEUNP group,and the difference was statistically significant.2.The results of HE staining showed that there were 11 cases of NEUNP and 16 cases of EOSNP.3.The results of immunohistochemistry showed that IDO1 was expressed in both nasal polyps and normal inferior turbinate tissues,and brown granules coul d be observed in the positive cells.Although IDO1 was expressed in EOSNP gr oup and NEUNP group,its staining intensity and expression quantity were signifi cantly lower than those in normal inferior turbinate tissue.H-score of IDO1 was24.16±8.54 in EOSNP group,13.76±7.96 in NEUNP group,and 65.44±18.11in control group respectively.The expression of IDO1 in the EOSNP group wa s lower than that in the NEUNP group,and the expression in the control group was significantly higher than that in the other two groups(P<0.05).The expressio n of IDO1 in NEUNP was lower than EOSNP(P<0.05).The expression of IDO1 protein in EOSNP,NEUNP and control group was detected by Western-blotting method.The results showed that compared with NEUNP group and EOSNP group,the expression level of IDO1 protein in cont rol group was higher,P<0.01,the difference was statistically significant.The exp ression level of IDO1 protein in EOSNP group was significantly higher than that in NEUNP group(P<0.05).4.The tissues of the three groups were detected by RT-qPCR method,and theΔCt values of EOSNP group,NEUNP group and control group were analyze d by analysis of variance.The results showed that theΔCt values of the three g roups were 2.37±0.44,3.40±1.19,1.39±0.78,12.3 and P<0.01 respectively.Th e results showed that the value of IDO1 mRNAΔCt in two groups of polyps w as significantly higher than that in normal inferior turbinate tissues.Compared wi th NEUNP group,IDO1 mRNAΔCt of EOSNP group was higher than that of EOSNP group,and the difference was statistically significant.ConclusionThe expression of IDO1 in nasal polyps is down-regulated,and the expressi on in NEUNP is lower than that in EOSNP,suggesting that IDO1 can be used as a potential new therapeutic marker for CRSwNP.
