Objective:The expression of S100 calcium binding protein A11(S100A11)mRNA and protein in pancreatic cancer tissues was observed,and the correlation between S100 calcium binding protein A11 mRNA and protein and clinical characteristics and prognosis of pancreatic cancer patients was analyzed.Methods:1.Through Oncomine database and Kaplan-Meier database,the differences of S100A11 mRNA expression in normal pancreatic tissues and pancreatic cancer tissues were statistically analyzed,and the correlation between its expression level and prognosis of pancreatic cancer patients was discussed.2.From March 1,2018 to August 31,2020,42 patients with pancreatic cancer who were admitted to the Second Department of Hepatobiliary and Pancreatic Surgery of the First Hospital of Jilin University and were diagnosed by pathology were collected.Immunohistochemistry,IHC)was used to detect the expression of S100A11 protein in pancreatic cancer.Image-Pro Plus 6.0 software was used to analyze protein expression.3.SPSS 23.0 statistical software was used to analyze the data.The relationship between S100A11 protein expression level and clinical data of pancreatic cancer patients was tested by Chi-square test of cross-table method.Kaplan-Meier method and log rank test were used to analyze the relationship between clinical information,S100A11 protein expression and total survival time and tumor-free survival time of pancreatic cancer patients.COX regression model was used to analyze the independent risk factors affecting the overall survival time of pancreatic cancer patients,and the difference was statistically significant(P < 0.05).Results:1.The analysis of Oncomine database showed that the expression level of S100A11 mRNA in pancreatic cancer tissues was significantly higher than that in normal pancreatic tissues(p < 0.05).Kaplan-Meier database analysis showed that the overall survival rate of pancreatic cancer patients with high S100A11 mRNA expression was lower than that of those with low S100A11 mRNA expression(P <0.05).2.By observing the immunohistochemical staining of pathological sections of patients with pancreatic cancer,it can be concluded that the expression rate of S100A11 protein in patients with preoperative CA125≤35 U/ml is lower than that in patients with preoperative CA125 > 35 u/ml(p<0.05).Compared with patients with pancreatic duct diameter ≥3 mm,the expression rate of S100A11 protein in patients with pancreatic duct diameter < 3 mm was higher(p < 0.05).There was no significant difference between S100A11 protein expression level and onset age,tumor size,lymph node metastasis number,preoperative CA199,pancreatic texture,preoperative blood glucose and pathological classification(P > 0.05).3.According to Kaplan-Meier univariate analysis of prognosis,the overall survival rate of pancreatic cancer patients with tumor diameter ≥3.5 cm was lower than that of patients with tumor diameter < 3.5 cm(P < 0.05).The total survival time of pancreatic cancer patients with high expression of S100A11 protein in tumor tissue was less than that of patients with low expression(P < 0.05).The age of onset,pancreatic texture,preoperative blood glucose,the number of lymph node metastasis,preoperative CA125,preoperative CA199 and pathological classification had no significant correlation with the overall survival rate(P > 0.05).COX multivariate analysis showed that high expression of S100A11 protein and tumor diameter ≥3.5cm were independent risk factors affecting the prognosis of pancreatic cancer.4.By Kaplan-Meier univariate analysis of prognosis,it was found that the tumor-free survival time of patients with low,medium and low differentiation of pancreatic cancer was shorter than that of patients with differentiated pancreatic cancer(P < 0.05).Patients with pancreatic cancer with high expression of S100A11 protein had less tumor-free survival time than those with low expression(P < 0.05).COX multivariate analysis showed that high expression of S100A11 protein and poorly,moderately and poorly differentiated pancreatic cancer tissues were independent risk factors affecting tumor-free survival of pancreatic cancer patients.Conclusions:1.The mRNA and protein levels of S100A11 are closely related to the occurrence and development of pancreatic cancer.The high level of S100A11 mRNA is a risk factor affecting the tumor-free survival time and overall survival time of pancreatic cancer patients,and the high expression of S100A11 protein is a risk factor affecting the overall survival time of pancreatic cancer patients.2.The high expression of S100A11 protein was negatively correlated with preoperative CA125 level and pancreatic duct diameter in patients with pancreatic cancer.3.Tumor ≥ 3.5 cm and high expression of S100A11 are the risk factors affecting the survival time of pancreatic cancer patients.The high expression of S100A11 is related to the tumor-free survival of pancreatic cancer patients.4.Low,medium-low differentiated pancreatic cancer and high expression of S100A11 are independent risk factors affecting tumor-free survival of pancreatic cancer patients.
