Effect Of Gastrodia Elata Alcohol Extract On The Improvement Of Neurological Function In Rats With Multiple Cerebral Infraction And Its Mechanism

Objective:With SPF health adult male SD rats as the research object,through the establishment of microsphere embolism caused by multiple cerebral infarction(MCI)animal model simulating clinical disease of MCI pathology observation of gastrodia elata alcohol extract of MCI model improvement of neurological function in rats,the mechanism of action was also discussed.Methods:The experimental animals were randomly divided into Sham operation group(Sham,pure water),Model group(Model,pure water),butylphthalide group(NBP,54mg/kg),Gastrodia elata pure extract low-dose group(Low-TM,4.95mg/kg),Gastrodia elata pure extract medium-dose group(Mid-TM,9.9mg/kg),Gastrodia elata pure extract high-dose group(High-TM,19.8mg/kg).Behavioral tests(Longa5 scale score,balance beam walking test,rope climbing test and gait analysis test)were used to evaluate the changes of impaired neurological function in model rats.Enzyme-linked immunosorbent assay was used to detect the effect of Gastrodia elata alcohol extract on serum BDNF content of model rats.The 5-bromo-2’-deoxyuridine(Brd U)labeling method was used to investigate the effects of Gastrodia elata alcohol extract on neurogenesis from ependymal down to ischemic cortex of lateral ventricle in model rats.Western blot was used to detect the effects of Gastrodia elata alcohol extract on the expression levels of Wnt/β-catenin signaling pathway related proteins in the cerebral cortex of model rats.Results:The results showed as follows:1.Gastrodia elata alcohol extract can promote the recovery of neurological function in MCI model rats.2.Gastrodia elata alcohol extract can promote the proliferation of neural stem cells in SVZ region of MCI model rats.3.Gastrodia elata alcohol extract can increase the number of neuroblasts in SVZ(Z1),corpus callosum(Z2)and ischemic injury(Z3)in MCI model rats,and promote their migration along the corpus callosum to the ischemic cortex.4.Gastrodia elata alcohol extract can increase the number of new neurons in the ischemic area of MCI model rats.5.Gastrodia elata alcohol extract can increase BDNF content in serum of MCI model rats.6.Gastrodia elata alcohol extract can increase the expression of Wnt3 A and TCF4,increase the ratio of p-GSK3β/GSK3β,and decrease the ratio of p-β-catenin/β-catenin in the cortex and brain tissue of MCI model rats.Conclusions:Gastrodia elata alcohol extracts may be through the activation of Wnt/β-Catenin signaling pathway to promote SVZ area neural stem cell proliferation after ischemic stroke,by raising the expression of BDNF in the serum raise more of endogenous nerve mother cells migrate to the damaged area,and differentiate into mature neurons,thus improving neural functional recovery of chronic cerebral ischemia mice.
