Objective: To establish a method to determine the contents of nine steroidal saponins and fifteen amino acids in rhizome and fibrous root of Paris polyphylla var.yunnanensis,evaluate the quality of wild and cultivated samples of P.polyphylla var.yunnanensis in Southwest China,study the correlation between various elements and the quality of P.polyphylla var.Yunnanensis,and explore inorganic elements which have an important impact on the contents of effective medicinal components.Methods: Contents of nine steroidal saponins and fifteen amino acids in rhizome and fibrous root of P.polyphylla var.yunnanensis from different habitats were determined by UPLC;the rhizome of P.polyphylla var.yunnanensis and soil samples were digested by microwave digestion instrument,and contents of inorganic elements were determined by ICP-MS;the forms of elements in rhizosphere soil were extracted by BCR sequential extraction and determined by ICP-MS;principal component analysis,cluster analysis and correlation analysis were applied to experimental data processing.Results: 1.Contents of polyphyllin Ⅰ and polyphyllin Ⅱ were higher than polyphyllin Ⅵ and gracillin in rhizome.Contents of polyphyllin Ⅵ,Ⅶ and H were rich in fibrous roots,while pseudo protodioscin,dioscin,slender dioscin and polyphyllin Ⅴ were not determined.2.Contents of aspartic acid,glutamic acid and arginine were higher than other types of amino acids in P.polyphylla var.yunnanensis samples from different producing areas.The average content of amino acids in cultivated P.polyphylla var.Yunnanensis samples was higher than that of natural crude drugs.3.Contents of each nutrient elements in rhizome and fibrous roots of P.polyphylla var.Yunnanensis in ranking were K > P > N,and contents of the three nutrient elements in rhizome were higher than that in fibrous roots.4.The characteristic inorganic elements in the rhizome of P.polyphylla var.Yunnanensis were Al,Fe,Zn,Mn,Co and Se,while characteristic inorganic elements in the rhizosphere soi were Mg,Al,Se,Ca and Mo.5.The major existing form of the fifteen inorganic elements in rhizosphere soil of P.polyphylla var.yunnanensis was residual form.Contents of water-soluble and exchangeable Ca,Mg and K were higher than other elements,and contents of P,B and Mo were extremely low;contents of Fe-Mn oxidative and organic Mg,Al,Ca,Fe and Mn were relatively high.6.Contents of available Mn and Fe in P.polyphylla var.yunnanensis samples from different habitats were rich.Contents of available Mn,Fe and Mg in rhizosphere soil from the wild production area were higher than those from cultivation areas,while contents of available Cu,Zn,Ca,and B were lower than those from cultivation areas.Conclusion: In this study,method was established to determine contents of saponins and amino acids in rhizome and fibrous root of P.polyphylla var.Yunnanensis by using UPLC.The quality of wild and cultivated samples were evaluated.The cultivated P.polyphylla var.Yunnanensis could be used as medicine instead of wild products,which was beneficial to protect the resources of natural P.polyphylla var.yunnanensis.The fibrous roots of P.polyphylla var.yunnanensis were rich in effective medicinal components,which couble be reused as raw materials.In addition,we found that the contents of Cu,Zn and Ni in rhizome and soil of P.polyphylla var.Yunnanensis were relatively high,the rhizome had a certain capbility of enriching Cu and Zn elements in soil and depleting Na,Al,Fe,B,Se and Sb.Besides Co,Se and Sb elements in soil had a significant correlation with contents of saponins in P.polyphylla var.Yunnanensis. |
