| Objective: To explore the effect of Huazheng Sanji prescription(HZSJ)combined with cisplatin(DDP)on the proliferation and migration of Hep G2 liver cancer cells induced by hypoxia,and the HIF-1α and VEGF proteins.Methods: Hep G2 liver cancer cells were divided into blank control group,hypoxic model group,DDP group,HZSJ group,HZSJ+DDP group,and the hypoxic environment was induced by the Co Cl2 method.MTT method detects cell proliferation ability,scratch test detects cell migration ability,immunofluorescence method and western blot detect HIF-1α and VEGF protein expression.Results: 1.Huazheng Sanji prescription can inhibit the proliferation of Hep G2 cells,and has obvious concentration-dependent;the combined application of Huazheng Sanji prescription and cisplatin inhibits cell proliferation significantly stronger than that of Huazheng Sanji prescription group(P<0.01)and the cisplatin group(P<0.01,P<0.001).2.High-dose Huazheng Sanji Decoction can inhibit the migration ability of Hep G2cells(P<0.01);the combination of Huazheng Sanji Decoction and cisplatin can significantly reduce the migration rate of Hep G2 cells after induction(P<0.01,P<0.01),P<0.01).3.The results of immunofluorescence and western blot showed that: Compared with the blank control group,the expression levels of HIF-1α and VEGF protein in the model group were significantly increased(P<0.05,P<0.01,P<0.001);compared with the model group and cisplatin In comparison,the expression of HIF-1α protein in the high-dose Huazheng Sanji prescription group(200μg/m L)and the combination medication group was reduced(P<0.05,P<0.01,P<0.001),but there was no difference between the groups.Compared with the model group,the high-dose Huazheng Sanji prescription group(200μg/m L)can reduce the expression of VEGF(P<0.05);compared with the model group and the cisplatin group,the combination group can reduce the expression of VEGF(P< 0.01,P<0.001),and has obvious concentration dependenceConclusion: Huazheng Sanji prescription has the effect of inhibiting the proliferation and migration of Hep G2 hepatocellular carcinoma cells under hypoxia,which may be related to reducing the expression of HIF-1α and VEGF,and its increase in the anticancer sensitivity of the chemotherapy drug cisplatin may be related to the second It is related to the cooperative inhibition of VEGF expression. |
