Experimental Study On The Effect Of Acupoint Application On Gastrointestinal Hormones In Rats With Gastric Ulcer

Objective 1.Based on histopathological observation about the Pathological changes of gastric tissue in rats,it is proved that acupoint application can prevent gastric ulcer.2.To observe the effects of acupoint application intervention on changes in serum gastrin and plasma motilin content of rats,and to explore the effects of acupoint application therapy on gastrointestinal hormones of rats with gastric ulcer,so as to provide theoretical basis for the study of the biological mechanism of gastric ulcer and the clinical treatment of gastric ulcer with acupoint application.Methods 1 A total of 40 healthy SPF Wistar male rats were selected and randomly divided into 4 groups: blank group S1,model group S2,sham acupoint sticking group S3 and acupoint sticking group S4,with 10 rats in each group.Adaptive feeding was first given for 7 days.Group S1 was fed normally without any treatment.Rats in group S2 were fed normally and made model.In S3 group,Pishu point and Weishu point were used for 10 days of treatment intervention and modeling.Rats in S4 group were selected Pishu point and Weishu point for acupoint application treatment,for 10 days of therapeutic intervention and modeling.After treatment and intervention,rats in group S1 were fasted for 24 h without fetch-water immersion,and rats in groups S2,S3 and S4 were fasted for 24 h before fetchwater immersion to make gastric ulcer model.After modeling,eyeball blood and stomach tissue were collected from rats in each group under anesthesia.2.He staining was used to observe the pathological changes of gastric tissue,observe the shedding of epithelial cells,bleeding,hoofgland tissue swelling,inflammatory infiltration and other conditions of gastric tissue in rats,and observe and evaluate the different improvement of physiological and pathological conditions of gastric ulcer in rats by acupoint application on the premise of determining the success of gastric ulcer modeling.3.The changes of plasma motilin MTL and serum gastrin GAS in each group were detected by enzyme linked immunosorbent assay ELISA to determine the intervention effect of acupoint application on gastric ulcer model rats.Results1.HE staining of gastric tissue:Blade group S1: The morphology of gastric tissue cells was complete,the morphology of gastric mucosa was normal,the secretion was less,and there was no edema,hyperplasia,inflammatory infiltration and other phenomena.Gastric ulcer model group S2: a large number of inflammatory cell infiltration and exhumed epithelial cells were observed in the gastric tissue,accompanied by bleeding,edema,hyperplasia of hoofgland tissue,and congestion and thickening of gastric mucosa.S3 of the sham acupoint sticking group: some inflammatory cells infiltrated in the gastric tissue,some epithelial cells were shed,accompanied by edema and hyperplasia of hoofand tissue.In the acupoint application group,S4: the gastric tissue morphology was regular,with a small amount of epithelial cells falling off and no connective tissue hyperplasia.Finally,it was determined that the gastric ulcer model in rats was successfully constructed and drug application treatment could effectively prevent gastric ulcer in rats.2.Changes of plasma motilin levels in rats with gastric ulcer:Compared with blank group S1,plasma motilin content in the other three groups was significantly increased P<0.05;Compared with model group S2,plasma motilin content of S4 rats in acupoint application group was significantly decreased P<0.01,the plasma motilin content in acupoint application group S4 was significantly lower than that in sham acupoint application group S3 P<0.05,suggesting that acupoint application can prevent gastric ulcer,regulate gastrointestinal peristalsis and reduce plasma motilin level.3.Changes of serum gastrin levels in rats with gastric ulcer:Compared with blank group S1,serum gastrin level of S3 rats in model group S2 and sham acupoint paste group was significantly decreased P<0.05;Compared with model group S2,serum gastrin level of S4 rats in acupoint application group was significantly increased P <0.05;Compared with S3 group,serum gastrin content of S4 rats in acupoint application group was significantly increased P<0.05,from the point of view of content,false point sticking can also stimulate rats and increase gastrin level,but acupoint sticking can significantly increase gastrin content,suggesting that acupoint sticking can improve serum gastrin level,inhibit the occurrence of gastric ulcer,and play a certain preventive role.Conclusion1.Water immersion-binding method can successfully create rat gastric ulcer model,and acupoint application can effectively prevent gastric ulcer in rats.2.acupoint application can effectively reduce plasma motilin content in gastric ulcer model rats;And significantly increased serum gastrin content.It is suggested that acupoint application can adjust gastrointestinal peristalsis,abnormal gastric acid secretion,repair gastric mucosa and activate the immune system.At the same time,the effect of acupoint application in the treatment of gastric ulcer may be realized by regulating gastrointestinal hormones,increasing the protective effect of cells and repairing gastric mucosa damage.
