Objective: To investigate the effect of GDF5 on proliferation and differentiation of dental pulp stem cells.Methods: Rat pulp cells were cultured in vitro,and cell phenotypes were identified by flow cytometry.Different concentrations of GDF5 and the addition of BMP2 were used to intervene the pulp stem cells.Cell proliferation and odontoblast differentiation ability were detected by CCK8,ALP and real-time quantitative PCR.Results: Rats dental pulp cells were isolated and identified by modified tissue enzymolysis.CCK8: Compared with the blank group,the two concentrations of GDF5 promoted the proliferation of pulp stem cells,the promotion effect of 1000ng/mL GDF5 was more obvious(P<0.05),and 100ng/mL BMP2 promoted the proliferation of pulp stem cells compared with the blank group(P<0.05).100ng/mL GDF5+100ng/mL BMP2 and1000ng/mL GDF5+100ng/mL BMP2 promoted the proliferation of pulp stem cells compared with the blank group.1000ng/mL GDF5+100ng/mL BMP2 promoted the proliferation of dental pulp stem cells less than that of 100ng/mL GDF5+100ng/mL BMP2(P<0.05).ALP: Compared with the blank group,both concentrations of GDF5 inhibited ALP activity,of which 1000ng/mL GDF5 inhibited ALP activity more obviously(P<0.05),100ng/mL BMP2 increased ALP activity compared with the blank group(P<0.05).100ng/mL GDF5+100ng/mL BMP2 and 1000ng/mL GDF5+100ng/mL BMP2 increased ALP activity compared with blank control group.1000ng/mL GDF5+100ng/mL BMP2 had a weaker effect on ALP activity than 100ng/mL GDF5+100ng/mL BMP2(P<0.05).PCR: Compared with the blank group,the two concentrations of GDF5 inhibited the odontogenic differentiation of pulp stem cells.1000ng/mL GDF5 inhibited the odontogenic differentiation of pulp stem cells more obviously(P<0.05),100ng/mL BMP2 promoted the odontogenic differentiation of pulp stem cells compared with the blank group(P<0.05).100ng/mL GDF5+100ng/mL BMP2 and 1000ng/mL GDF5+100ng/mL BMP2 promoted odontogenic differentiation of pulp stem cells compared with blank group.1000ng/mL GDF5+100ng/mL BMP2 had weaker effect on odontogenic differentiation than 100ng/mL GDF5+100ng/mL BMP2(P<0.05).Conclusions:Both100ng/mL and 1000ng/mL of GDF5 promoted the proliferation of pulp stem cells but inhibited their odontogenic differentiation.100ng/mL BMP2 promoted the proliferation and odontoblast differentiation of dental pulp stem cells. |
