ObjectiveIn this study,the expression of gelsolin(GSN)in patients with early breast invasive ductal carcinoma with lymph node metastasis and patients without lymph node metastasis was analyzed by immunohistochemical method,and its effect on the proliferation of breast cancer MCF-7 cells was preliminarily investigated.As well as the impact of invasion,the corresponding molecular mechanism is analyzed at the same time.MethodsA total of 40 female patients with invasive ductal carcinoma of the breast who were diagnosed in the Third Affiliated Hospital of Jinzhou Medical University and the First Affiliated Hospital of Jinzhou Medical University from January 2018 to December 2018 were selected and collected,and then their cancer tissue samples were obtained.In the experiment,a total of 20 cases of T1N1-3stage and 20 cases of T2-3N0stage were collected.At the same time,20normal tissue samples of breast fibroadenoma patients were collected as controls.Immunohistochemistry was used to detect the expression of GSN.According to the research needs,complete the construction of the GSN overexpression vector,and realize the transfection into breast cancer MCF-7cells after completion.The transfection efficiency was detected by RT-q PCR and verified by Western blot.CCK8 method and Transwell test were used to detect the proliferation and invasion of transfected cells.In order to clarify its mechanism,Western blot was used to detect the effect of GSN overexpression on the PI3K-AKT signaling pathway in breast cancer MCF-7 cells.Results1.Compared with normal tissues adjacent to breast fibroadenoma,GSN is expressed at a low level in both T1N1-3and T2-3N0breast cancer tissues,which is significantly lower than that of normal tissues surrounding breast fibroadenoma,and is similar to breast cancer T2-3N0Compared with tissues,GSN content was the lowest in T1N1-3breast cancer tissues(P<0.05).2.RT-q PCR and Western blot detection showed that compared with the normal control and the empty vector group,after the overexpression vector GSN-pc DNA3.1 was transfected into MCF-7 cells,the overexpression of GSN significantly increased the relative expression levels of GSN m RNA and protein.(P<0.05).3.The results of CCK8 test showed that compared with the normal control group,the MCF-7 cells were affected by the GSN overexpression vector transfected into the MCF-7 cells,and their proliferation was significantly inhibited(P<0.05).4.The results of the Transwell test showed that,compared with the normal control and the empty vector group,the MCF-7 cells were affected by the GSN overexpression vector transfected into the MCF-7 cells,and the specific manifestation was that the invasion was significantly inhibited(P<0.05).5.Western blot results showed that compared with the normal control group,under the background of GSN overexpression,the down-regulated PI3K and p-AKT proteins in the PI3K-AKT signaling pathway can be found after detection(P<0.05).Conclusions1.GSN may be related to lymph node metastasis of breast invasive ductal carcinoma.2.GSN may inhibit the expression of PI3K and p-AKT protein in MCF-7cells,thereby preventing the proliferation and invasion of breast invasive ductal carcinoma cancer cells. |
