ObjectiveTo explore the effects of baicalein on the proliferation,migration and apoptosis of ovarian cancer Skov3 cells,and explore its possible molecular mechanisms.Methods1.Treat ovarian cancer Skov3 cells with different concentrations of baicalein,observe the changes in the proliferation ability of ovarian cancer Skov3 cells by MTT method,and screen out the concentration of baicalein used in subsequent experiments.2.Use the clone formation experiment to observe the changes in theclone formation ability of ovarian cancer Skov3 cells after treatment with the experimental concentration of baicalein.3.The cell scratch test was used to detect the changes in the migration and healing ability of ovarian cancer Skov3 cells after treatment with the experimental concentration of baicalein.4.Use flow cytometry to observe the changes in ovarian cancer Skov3 cell apoptosis after treatment with experimental concentration of baicalein.5.Western blot was used to observe the changes in the expression of apoptosis and migration-related proteins in ovarian cancer Skov3 cells after treatment with experimental concentrations of baicalein.6.Western blot was used to detect the changes in the expression of related proteins in the NF-κB pathway after the experimental concentration of baicalein was treated.7.The immunofluorescence experiment was used to observe the changes of NF-κBp65 expression in ovarian cancer Skov3 cells after treatment with baicalein at the experimental concentration.Results1.After different concentration gradients of baicalein act on ovarian cancer Skov3 cells for 24 h,48h,72 h,the proliferation of ovarian cancer cells is obviously inhibited and is concentration-dependent,and its IC50 is respectively 85.16μmol/L、83.53μmol/L、83.04μmol/L.2.After 10 days of treatment of ovarian cancer Skov3 cells with the experimental concentration of baicalein,the clone formation experiment showed that the cell clone formation rate of each drug-added group was significantly decreased compared with the control group,and it was concentration-dependent.3.After ovarian cancer Skov3 cells were treated with baicalein at the experimental concentration for 24 h and 48 h,the cell scratch test showed that the cell migration and healing ability of the drug-added group was significantly reduced compared with the control group.4.After the experimental concentration of baicalein acted on ovarian cancer Skov3 cells for 24 hours,the results of flow cytometry showed that compared with the control group,the apoptosis rate of the drug-added group was significantly increased.5.After the experimental concentration of baicalein interferes with ovarian cancer Skov3 cells for 24 hours,the western blot results showed that the expression of cell migration-related proteins N-Cadherin and Vimentin in the drug-added group was significantly lower than that in the control group.At the same time,compared with the control group,the expression of apoptosis-related proteins bax and cleaved caspase-3 increased,and the expression of bcl-2 decreased significantly.Compared with the control group,the expression of apoptosis protein inhibitor c-IAP1 decreased,while the expression of pro caspase-9 decreased,and the expression of cleaved caspase-9 increased.6.After ovarian cancer Skov3 cells were treated with baicalein at an experimental concentration,western blot results showed that the expression of NF-κB pathway related proteins p NF-κBp65 and p IκB-α was significantly lower than that of the control group,and the expression of IκB-α was significantly higher than that of the control group.7.The results of immunofluorescence experiments showed that after baicalein treating ovarian cancer Skov3 cells for 24 hours,compared with the control group,the nucleus of P65 protein increased in the cytoplasm.ConclusionBaicalein can inhibit the proliferation and migration of ovarian cancer Skov3 cells and promote their apoptosis.Its inhibitory effect on the migration of ovarian cancer Skov3 cells may be related to the regulation of the expression of migration-related proteins,its mechanism of promoting the apoptosis of ovarian cancer Skov3 cells may be related to the expression of apoptosis-related factors regulated by the NF-κB signaling pathway.
