ObjectiveTo analyze the expression and correlation of micro RNA-126(mi R-126)and vascular endothelial cell adhesion molecule-1(VCAM-1)in placentas of patients with early-onset preeclampsia(EOPE),as well as the effects of the two on trophoblast invasion,and to explore the role and their relationship in the pathogenesis of EOPE.MethodsWe selected 60 pregnant women who delivered in the Third Affiliated Hospital of Zhengzhou University from November 2019 to may 2020 as the study objects.The placenta tissue after delivery was collected as the experimental materials.Among them,30 placenta tissues from EOPE patients were named PE group,and 30 healthy pregnant women with normal group were named Normal group.We further cultured and transfected HTR-8/SVneo cells to study the effects of mi R-126 and VCAM-1 on the invasion of trophoblast.Immunohistochemistry was used to locate VCAM-1 in the placental tissue,and quantitative real-time PCR(RT-PCR)was used to detect expression of mi R-126 and VCAM-1 m RNA in the placental tissue of the two groups of women,and the correlation between them was analyzed.Western blot(WB)method was used to detect the expression of VCAM-1 protein in the placental tissues of the two groups.Then we observed the invasive ability of HTR-8/SVneo transfected with mi R-126 by Transwell invasion test,and detected the expression of mi R-126 and VCAM-1 in HTR-8/SVneo transfected with mi R-126 mimics,mi R-126 inhibitor and negative control by RT-PCR and WB.Graph Pad Prism 8.0 and Image J were used for image processing and editing,and SPSS25.0 software was used for statistical analysis.P<0.05,the difference was statistically significant.Results1.The average age and the average number of pregnancies of patients with early-onset preeclampsia were lower than those of the normal group,and the body mass index(BMI)before pregnancy was higher than that of the normal group,with no statistical significance(P>0.05).The average gestational age,neonatal weight and placental weight of patients with early-onset preeclampsia were lower than those of the normal group,with statistical significance(P<0.01).2.In placental tissue,VCAM-1 was mainly expressed in trophoblast cell membrane and cytoplasm.Compared with the normal group,the expression level of mi R-126 in PE group was higher,while the m RNA and protein levels of VCAM-1were lower,and the difference were statistically significant(P<0.01).3.Correlation analysis showed that the expression of VCAM-1 and mi R-126 in the placental tissues of the two groups was negatively correlated(r=-0.391,P<0.05).4.Transwell invasion assay results showed that the invasion ability of HTR-8/SVneo cells transfected with mi R-126 mimics decreased,while the invasion ability of HTR-8/SVneo cells transfected with mi R-126 inhibitor increased.5.In vitro cell experiment results showed that the expression level of VCAM-1 in the mi R-126 mimics transfection group was lower than that in the mi R-126 mimics negative control(NC)group,and the expression level of VCAM-1 in the mi R-126 inhibitor transfection group was higher than that in the mi R-126 inhibitor NC group.ConclusionIn placental tissues of patients with early-onset preeclampsia,mi R-126 may be involved in the pathogenesis of early-onset preeclampsia by negatively regulating the expression of VCAM-1 and inhibiting the invasion ability of trophoblast cells. |
