| BackgroundColorectal cancer(CRC)poses a serious public health threat due to its high recurrence rate and low 5-year survival rate.CircRNAs have emerged as promising molecules in the area of oncology clinical research in different malignancies,including CRC.However,the regulatory effect and clinical value of circRNA 1268 on CRC remain unclear.PurposeTo explore the biological role of circ1268 in CRC,and its potential value in the prognosis evaluation and clinical treatment of CRC patients,in order to discover novel biological markers for the early diagnosis,therapeutic intervention and prediction of prognosis of CRC.MethodsQuantitativeReal-time fluorescent PCR(qRT-PCR)was used to detect the expression level of circ1268 in 66 pairs of colorectal cancer tissue samples and adjacent tissue cells collected in our hospital.Kaplan-Meier test,Log-rank test and Cox regression analysis were used to predict the potential prognostic value of circ1268 in CRC patients.In addition,the expression of circ1268 in colon cancer cells HCT116 was knocked down by siRNA interference technology,and the effect of circ1268 molecule on the proliferation of HCT116 cells was detected by CCK-8 assay.The effect of CIRC 1268 on HCT116 cell invasion was investigated in vitro by Transwell assay.ResultsThe expression level of circRNA 1268 in 66 pairs of colorectal cancer tissues and paracancerous tissues was detected by qRT-PCR.The results showed that the expression level of circRNA 1268 in colorectal cancer tissues was significantly lower than that in paracancerous normal tissues.The colorectal cancer patients in this study were divided into circRNA 1268 low expression group and high expression group.Kaplan-Meier analysis showed that the disease-free survival(DFS)and overall survival(OS)of CRC patients with low circ1268 expression level were lower than those with high circ1268 expression level(P<0.05).Statistic analysis of clinical data and circ1268 relative expression level showed that circ1268 was an independent prognostic factor in CRC patients.In addition,in vitro cytology experiments confirmed that the interference of circ1268 expression increased the proliferation and invasion ability of HCT116 cells.ConclusionThe expression of circ1268 is down-regulated in colorectal cancer,and the expression level of circ1268 is an independent factor affecting the prognosis of CRC patients.As a non-coding RNA molecule that inhibits the proliferation and invasion of colorectal cancer cells,circ1268 negatively regulate the progression of CRC,and is expected to become a ideal prognostic marker and a novel target for biological therapy in CRCpatients. |
