Coreopsis tinctoria Nutt.,the other name is Coreopsis basalis,is a rare alpine plant.It has the obvious prevention and the treatment effect to a variety of diseases.With the development of science and technology,it is analyzed that the main contents of C.inctoria are composed of polysaccharides,flavonoids,volatile oil,phenylpropanoids,aikyne,pigments,microelements and so on.By modern pharmacological researches,the active ingredients of Coreopsis tinctoria are complex,including hypotensive,hypolipidemic,hypoglycemic,antioxidant,antibacteria and anti-inflammation.It is widely used in medicine,food,health care products,cosmetics and other fields.For the past few years,industrial cultivation of C.tinctoria Nutt.has been carried out as a kind of tea and health care products in parts of xinjiang,China.In this study,the content of total flavonoids of C.tinctoria from three main origins which are Pishan county Keliyang township,Changji jimsar county and grape ditch in Turpan was determined,and the contents of chlorogenic acid,3,5-O-caffeoylquinic acid and galuteolin in C.tinctoria were compared with the contents in Chrysanthemum morifolium Ramat cv.’Hangju’.The antioxidant activity,antibacterial activity and anti-inflammatory activity were determined to analyze the differences in the functonal activity of C.tinctoria from different origins at different altitudes.Main results and conclusions of the study are as follows:(1)The total flavonoids of C.tinctoria and Hangju were determined by uv-vis spectrophotometry;the contents of chlorogenic acid,3,5-O-caffeoylquinic acid and galuteolin were determined by the UPLC method.The results showed that there were similar major active ingredients in C.tinctoria and Hangju,but there were significant differences in contents.The content of flavonoids,chlorogenic acid,3,5-O-caffeoylquinic acid and galuteolin in Hangju were 61.94 mg·g-1、5.33 mg·g-1、14.76 mg·g-1、9.22 mg·g-1.The content of flavonoids,chlorogenic acid,3,5-O-caffeoylquinic acid and galuteolin in C.tinctoria from three origins were between 186.59~278.68 mg·g-1,1.34~2.62 mg·g-1,3.59~8.63 mg·g-1 and 29.91~33.93 mg·g-1.In addition to galuteolin,the content of other components increased with the elevation of producing area.The contents of main active components in different samples were significantly different.(2)The antioxidant activity of C.tinctoria from three origins were evaluated by the capacity of scavenging the DPPH radical,superoxide anion free radical,hydroxyl radical and ABTS radical and iron ion reduction capacity method.The results showed that C.tinctoria from three origins had a good reduction capacity of iron ion and it also had the certain scavenging effects on DPPH free radicals,superoxide anion free radical,hydroxyl radical and ABTS radical.When the mass concentration was between 0.1~2.0 mg·mL-1,the antioxidant activity of C.tinctoria increased with the increase of concentration.By analysis,the order of free radical scavenging capacity was Kiliyang>Changji>Turpan and the content of total flavonoids in C.tinctoria was positively correlated with its antioxidant ability.The scavenging capacity of C.tinctoria was lower than that of vitamin C in superoxide anion free radical and hydroxyl radical,and the scavenging capacity of DPPH free radicals and ABTS free radicals was equivalent to that of vitamin C.(3)The antibacteria activity of C.tinctoria from three origins were evaluated by filtering paper method.At the same time,the minimal inhibitory concentration was determined.The results showed that C.tinctoria from three different origins had a good antibacterial activity.The order of the antibacterial activity of C.tinctoria was staphylococcus aureus>bacillus subtilis>salmonella>escherichia coli;the order of the antibacterial activity of C.tinctoria from strong to weak was Keliyang>Changji>Turpan.According to the correlation coefficient analysis,the ability toinhibit four strains was found positively correlation with the content of total flavonoids in C.tinctoria.The bacteriostatic experiment results showed that the minimum bacteriostatic concentrations(MIC)for various microbes were as follows:Escherichia coli and Salmonella enteritis were 0.625 mg·mL-1,Staphylococcus and Bacillus subtilis were 0.3125 mg·mL-1..(4)In order to study the anti-inflammatory activity in vitro of C.tinctoria from different origins,the anti-xanthine oxidase activity was taken as the index,meanwhile,allopurinol was used as positive control.The inhibiting effect to xanthine oxidase of total flavonoids in C.tinctoria was determined,and the results showed that the inhibitory capacity increased with increasing of mass concentration.In equal mass concentration,the inhibition rate of xanthine oxidase of C.tinctoria from different origins was Keliyang>Changji>Turpan.By calculation,the half-inhibitory concentrations(IC50)of C.tinctoria from three different origins were determined to be 0.34 mg·mL-1(Keliyang),0.44 mg·mL-1(Changji),0.48 mg-mL-1(Turpan).The results suggested that the content of total flavonoids in C.tinctoria was negative correlation to the IC50 for xanthine oxidas,but the correlation was not significant.The inhibition mechanism of C.tinctoria showed reversible inhibition and the extract of C.tinctoria was weak synergies. |
