| Dactylicapnos torulosa(Papaveraceae)can clear away heat and toxin,reduce inflammation and pain,reduce blood pressure.This work studied the chemical constituents on the petroleum ether,dichloromethane and n-butanol fractions from the 95%ethanol extract of D.torulosa,using multiple separation techniques including silica gel chromatography,reverse phase liquid chromatography and high speed counter current chromatography.Thirty-five compounds were identified by nuclear magnetic resonance,circular dichroism and IR spectra,as well as comparison with literatures.The compounds include 11 protoberberine alkaloids,5 protopine alkaloids,6 benzophenanthridine alkaloids,4 amides,2 terpenes,3 flavonoids,3 coumarins and one sugar.Among them,two were new compounds,i.e.alkaloids 2-3.And 27 compounds were isolated from D.torulosa for the first time.The anti-inflammatory,antithrombotic,antiplatelet aggregation and anti MPTP induced Parkinson’s disease activities of some isolated compounds from D.torulosa were studied.It was found that protopine alkaloids 4 and 6 had the best antithrombotic effect,and anti-thrombolic effectcould reach 100%at the concentration of 50 μM.For compounds 7,phenanthridine alkaloids 1,8,10,anti-thrombolic effects at 50 μM were 81.3%,79.4%,84.4%,79.3%,respectively,showing strong antithrombotic activity.Coumarin 33,flavones 31,32,phenanthridine alkaloids 1,8,10 and protoberberine alkaloid 15 had obvious anti-inflammatory effect,while triterpenes 27,28 had reletaively weak anti-inflammatory effect;protopine alkaloids showed anti-inflammatory activity at high concentration as 25 μM.In the test of dopaminergic neuron protective activity using Parkinson’s disease Model,compound 1 showed obvious neuroprotective activity,with the dopaminergic neuron length ratios of 1.41,1.28 and 1.13 at 5,10 and 50 μM,respectively,and it also prevented the MPTP induced Parkinson-like action in behavioral experiments.This work will be helpful for future research on D.torulosa and the genus Dactylicapnos. |
