Objective: To explore the possible mechanism of Shuangyu Tiaozhi Granules intervention in the reduction of brain injury and the recovery of nerve function after ischemic stroke,and to provide the characteristic research and clinical application of the treatment of ischemic stroke with the method of turbid turbidity New experimental support.Methods: A rat model of middle cerebral artery occlusion(MCAO)was established.In this study,high-and low-dose Shuangyu Tiaozhi Granules or butylphthalide were administered intragastrically to intervene in the MCAO model of rats.The crosswalk test(BWT)was used to evaluate the recovery of neurological function in each group of rats;HE staining was used to observe the pathological changes of brain tissue in the infarct area;Nissl staining was used to observe the changes of neurons in the infarct area;Using RT-PCR technology,Western blot,immunofluorescence detection of neurite outgrowth factor A(Nogo-A),myelin associated glycoprotein(MAG),oligodendrocyte myelin glycoprotein(OMgp),Nogo receptor(NgR),protein kinase C(PKC),Rho A relative expression of mRNA or protein;immunoenzyme linked method(ELISA)was used to determine the expression of TNF-α and IL-6 in serum.Results: According to the research results,after the intervention of Shuangyu Tiaozhi Granules,the neurological deficit of MCAO model rats improved,and peripheral inflammatory response and apoptotic cells decreased.Further research shows that Shuangyu Tiaozhi Granules could significantly reduce the expression of Nogo-A,MAG,OMgp and NgR,and down-regulate the expression level of PKC / RhoA signaling pathway.Moreover,the effect of Shuangyu Tiaozhi Granules on reducing brain damage in MCAO model rats is dose-dependent.These data indicate that Shuangyu Tiaozhi Granules can reduce brain damage after ischemic stroke and can be used as a candidate drug for the treatment of ischemic stroke.
