Fritillariae Thunbergii Bulbus,as the dry bulb of Fritillaria thunbergii Miq.,a plant of Liliaceae,has the effects of anti-inflammation,clearing heat,resolving phlegm and relieving cough,detoxification,dispelling knot and eliminating carbuncle.In this paper,taking Fritillariae Thunbergii Bulbus as the research object,the fingerprints and anti-inflammatory and expectorant experiments of Fritillariae Thunbergii Bulbus from different habitats were analyzed and studied.The quality markers of its anti-inflammatory and expectorant effects were preliminarily obtained,which provided a basis for the follow-up quality control of Fritillariae Thunbergii Bulbus.The main research contents were as follows:1.Analysis of chemical constituents of Fritillariae Thunbergii Bulbus.In this study,UPLC-Q-TOF-MS technique was used to study the fingerprints of Fritillariae Thunbergii Bulbusi from 10 habitats and identify their common peak chemical constituents under positive and negative ion mode.A total of 34 compounds were identified,such as Peimisine,Imperialine,Peiminine,Peimine,Oberidine,Caffeine,Picropodophyllotoxin,Zhebeine and Cyclopamine and so on.Through the induction and summary of the structural cracking pathway,the main cracking law was usually the loss of some small molecules such as H,CH3,H2O and so on,which provided data reference for follow-up experiments.2.Study on bioevaluation of Fritillariae Thunbergii Bulbus based on anti-inflammatory and expectorant efficacy model:Anti-inflammatory and expectorant experiments were carried out on ICR mice by intragastric administration of Fritillariae Thunbergii Bulbus from 10habitats.The results showed that the anti-inflammatory effects were as follows:Xingyi of Guizhou(Yuanzi),Zhangshui of Zhejiang,Anhua of Hunan(sulfur fumigation),Longguan of Zhejiang,Xingyi of Guizhou,Panan of Zhejiang,Zhoushan of Zhejiang,Southwest Guizhou,Nantong of Jiangsu,Panan of Zhejiang(sulfur fumigation),and the effect of high dose group was better than that of low dose group.The effects of resolving phlegm were Zhangshui of Zhejiang,Southwest Guizhou,Xingyi of Guizhou(Yuanzi),Nantong of Jiangsu,Anhua of Hunan(sulfur fumigation),Longguan of Zhejiang,Xingyi of Guizhou,Panan of Zhejiang,Panan of Zhejiang(sulfur fumigation)and Zhoushan of Zhejiang.The efficacy of high dose group was better than that of low dose group.3.Preliminary discovery of quality markers of Fritillariae Thunbergii Bulbus.Bivariate correlation analysis and grey correlation analysis were used to study the correlation between the spectral effects.Finally,the chemical components related to the efficacy were obtained.The results showed that there were 9 chemical constituents related to anti-inflammation,namely:Peiminine,Peimine,cyclopamine,Eleutheroside A,polyphyllin V,Picropodophyllotoxin,Beta-Sitosterol,ent-kaur-15-en-17-ol,ent-17-norkauran-16-one.There were 6 chemical components related to expectorant action,which were adenosine,peimisine,imperialine,peiminine,peimine,and zhebeine.4.Determination of four components in Fritillariae Thunbergii Bulbus from different habitats:The contents of four active components(Peimisine,Imperialine,Peiminine,Peimine)in Fritillariae Thunbergii Bulbus from different habitats were determined by HPLC-ELSD.The results showed that the contents of peimisine from high to low were Southwest Guizhou,Xingyi of Guizhou,Zhangshui of Zhejiang,Panan of Zhejiang,Zhoushan of Zhejiang,Longguan of Zhejiang,Panan of Zhejiang(sulfur fumigation),Xingyi of Guizhou,Nantong of Jiangsu,Anhua of Hunan(sulfur fumigation),and the content was in the range of 0.186~1.1069mg/g.The contents of imperialine from high to low were Southwest Guizhou,Panan of Zhejiang,Panan of Zhejiang(sulfur fumigation),Zhoushan of Zhejiang,Zhangshui of Zhejiang,Anhua of Hunan(sulfur fumigation),Xingyi of Guizhou(Yuanzi),Longguan of Zhejiang,Nantong Jiangsu,Xingyi of Guizhou,and the content was in the range of 0.2047~1.2439 mg/g.The contents of peiminine from high to low were Southwest Guizhou,Xingyi of Guizhou,Panan of Zhejiang(sulfur fumigation),Panan of Zhejiang,Zhoushan of Zhejiang,Anhua of Hunan(sulfur fumigation),Zhangshui of Zhejiang,Longguan of Zhejiang,Xingyi of Guizhou,Nantong of Jiangsu,and the contents were in the range of 0.0842-0.6115 mg/g.The contents of peimine from high to low were Southwest Guizhou,Xingyi of Guizhou,Panan of Zhejiang(sulfur fumigation),Zhoushan of Zhejiang,Anhua of Hunan(sulfur fumigation),Zhangshui of Zhejiang,Panan of Zhejiang,Longguan of Zhejiang,Xingyi of Guizhou,Nantong of Jiangsu,and the contents were in the range of 1.4585 and 11.7696 mg/g.To sum up,this study preliminarily revealed the quality markers of anti-inflammation and expectoration of Fritillariae Thunbergii Bulbus,which provided a more reliable theoretical basis for the subsequent formulation of scientific and effective quality control standards and the development of Fritillariae Thunbergii Bulbus.
