| Objective:1.Establish stress loading model in vivo to identify the location of EMMPRIN expression in periodontal tissue under mechanical loading.2.To explore the possible relationship between EMMPRIN and vascular endothelial growth factor(VEGF)in periodontal tissue remodeling under mechanical stress.Materials and methods:1.Establish the experimental rat model of periodontal remodeling,and detect the alveolar bone remodeling of rats under different stress and different action time by Micro-CT.2.After decalcification,paraffin sections were made to detect the expression and localization of EMMPRIN and VEGF in periodontal tissues by chemical immunohistochemistry.3.To explore the role of EMMPRIN and VEGF in periodontal tissue remodeling under different stresses.Result:1.After stress loading,the first molars moved to the middle at different degrees.When loading 200 g,the alveolar bone in traumatic group appeared bone loss.According to variance analysis and Bonferroni test,the tooth movement distance in traumatic group(200g)was significantly greater than that in orthodontic group(P < 0.05),but there was no significant difference in the distance of tooth movement between 7 days and 14 days(P > 0.05).2.EMMPRIN was significantly positive in the gingiva and PDL of the tension side and compression side after stress(P < 0.05).The expression of EMMPRIN in PDL on the tension side was correlated with the force value,but not with the applied time.The expression of EMMPRIN in PDL on the compression side was correlated with the applied time when applied 200 g force,and there was no significant difference between 7 d and 14 d when applied 50 g force.3.The expression of VEGF in periodontal tissue was not significantly different from that in control group at 7 days after stress,but was significantly stronger at 14 days than that in control group.The expression of VEGF in PDL of tension side was stronger than that of compression side in each group.Conclusion:1.Mechanical stress can up-regulate the expression of EMMPRIN and VEGF in periodontal tissue.2.Under physiological and pathological conditions,both EMMPRIN and VEGF are involved in periodontal remodeling under stress. |
