| Objective: To investigate the expression and clinical value of PTN in breast cancer.Methods:Blood samples of breast cancer patients(113 cases),benign breast lesions(32cases)and healthy volunteers(40 cases)which confirmed by postoperative pathology were collected,serum PTN levels in breast cancer patients,benign breast lesions and healthy subjects were detected by ELISA.Real-time PCR and immunoblotting Western blot)was used to detect the expression of PTN m RNA and protein in cancer tissues and adjacent tissues.At the same time,the expression levels of CA153,CEA and CA125 in plasma of patients with breast cancer,benign breast lesions and healthy subjects were determined by electrochemical luminescence,and the receiver operating characteristic curve was drawn and the diagnostic efficacy of PTN,CA153,CEA and CA125 for breast cancer was analyzed,as well as the value of the four combined tests.Results: The serum PTN expression level [573±164.3pg/ml] in breast cancer patients group is higher than that in benign breast lesions group [ 338.1±130pg/ml ]and healthy control group [296.6±86.5pg/ml](P<0.05).The differences between the benign lesion group and the healthy control group have no statistical significance(t=-1.56,P=0.125).And,the expression of PTN m RNA and protein in the cancer tissue was significant higher than the adjacent normal tissues(P<0.05).In addition,the expression of PTN in breast cancer patients was closely related to tumor metastasis,TNM stage and tumor differentiation degree(P<0.05).The AUC of PTN for the diagnosis of breast cancer was 0.913(95% CI 0.872-0.954,P<0.05)higher than serum CA153,CEA,CA125.The area under the receiver operating characteristic curve was the largest when the four tests were combined(AUC=0.99).Conclusion1.The expression of PTN in the peripheral blood of breast cancer patients was significantly higher than that of the benign breast lesions group and the healthy control group.Moreover,the clinicopathological features of tumor metastasis,tumor differentiation degree and TNM stage were closely related to the expression of PTN.Detection of serum PTN levels is helpful in assessing the risk of metastasis and prognosis of breast cancer.2.The expression of PTN m RNA and PTN protein in breast cancer tissue was significantly higher than that of the adjacent normal tissues.The result indicating that PTN expression is closely related to breast cancer.3.Combined detection of serum PTN,CA153,CEA and CA125 can significantly improve the diagnostic value of breast cancer. |
