Objective:The electromagnetic environment in human life has been changing dramatically with the development of industry and social progress.Excessive electromagnetic radiation can cause electromagnetic pollution and affect human health,which is one of the environmental pollution factors and that cannot be ignored.In our previous researches,we found that extremely low frequency electromagnetic fields(ELF-EMFs)can exert an influence on proliferation and secretion of some cytokines in RPE cells,which in turn affected the expression of MMP-2 and COLIA1.It may lead to the occurrence of myopia and puerarin reversed these adverse effects to some extent.Radio frequency electromagnetic radiation(RF-EMF)is also closely related to human life.However,there are few reports about the impact of radio frequency electromagnetic radiation on human eyes.Our present study explored whether a 900 MHz radio frequency electromagnetic radiation had an effect on the function of RPE cells as well as the prevention and treatmen of puerarin from the perspective of cells and molecules.Methods:RPE cells were cultured in vitro and divided into three groups.The radiation group was given a 900 MHz radio frequency radiation exposure and the control group was not given any exposure.The puerarin group was treated with a 900 MHz radio frequency radiation and puerarin simultaneously.Before formal experiments,the most appropriate radiation duration and puerarin treatment concentration were screened by two preexperiments.The proliferation of RPE cells was detected by the cell-counting kit-8(CCK8)assay.The expression of TGF-β2 mRNA,TGF-β2 protein and protein secretion of RPE cells were measured by Real-Time quantitative polymerase chain reaction(Real-Time PCR),Western blot and Enzyme-linked immunosorbent assay(ELISA),respectively.The detection methods of bFGF were the same as those of TGF-β2.Results:(1)It was found that the expression of TGF-β2 protein secretion of RPE cells began to increase after 6 hours of continuous irradiation and the concentration of puerarin at 200umol/L was the optimal therapeutic concentration by setting the radiation duration gradient and the puerarin treatment concentration gradient,respectively.(2)Compared with the control group,the viability of RPE cells in the radiation group was lowered.The level of TGF-β2 mRNA,TGF-β2 protein and protein secretion of RPE cells were up-regμLated under the exposure in contrast with the control group(P < 0.05).The level of bFGF mRNA,bFGF protein and protein secretion of RPE cells were down-regμLated under the exposure in contrast with the control group(P < 0.05).(3)Compared with the radiation group,the proliferation activity of RPE cells in puerarin group was increased.The level of TGF-β2mRNA,TGF-β2 protein and protein secretion of RPE cells treated with puerarin were decreased in contrast with the radiation group(P < 0.05).The level of bFGF mRNA,bFGF protein and protein secretion of RPE cells treated with puerarin were enhanced in contrast with the radiation group(P < 0.05).Conclusions: The 900 MHz radio frequency electromagnetic radiation may lead to some ocular diseases related with reduced proliferation activity of RPE cells by regulating the expression and release of TGF-β2 and bFGF.
