ObjectiveIn this paper,healthy male Kunming mice were used to prepare the renal injury model of cisplatin chemotherapy,and the effect of acupuncture on the kidneys of cisplatin chemotherapy mice was observed from the aspect of cell morphology by comparing the changes of renal histopathological in mice before and after acupuncture treatment,and the reactive oxygen species(ROS)in renal tissue of mice were detected by enzyme-linked immunoassay(ELISA).The content of Malondialdehyde(MDA)and superoxide dismutase(SOD)was preliminarily discussed based on oxidative stress response on the mechanism of acupuncture repair and improvement of renal injury caused by cisplatin.MethodAfter 3 days of adaptive feeding of 70 healthy male SPF mice,weigh,remove the poles and retain 60.The mice were divided into blank group(A),model group(B),acupuncture Group(C),moxibustion Group(D),15 in each group,dyeing number.Each group of mice was given by peritoneal injection at the dose of 0.01m L/g,and the blank group was given 0.9%Nacl solution,model group,acupuncture Group,moxibustion group to give 1mg/m L concentration of cisplatin solution,a total of 1doses,24 hours after the model success[1].Acupuncture Group and moxibustion group selected“Dazhui”,“Ganshu”(double),“Shenshu”(double),“Zusanli”(double)four points for treatment,acupuncture group using tubular injection method,each acupoint needle 3mm,static needle retention 6min;moxibustion Group every two acupoints for a group of moxibustion treatment,each group of Acupoint moxibustion 3min total 6min;1 times a day,Continuous treatment for 5 days,blank groups and model groups only accompanied by fixed,not treated.Before and after treatment,3 mice were randomly selected in each group,the kidneys were removed with 4%neutral polyformaldehyde fixation,renal histopathological observation was carried out,and the other mice used enzyme-linked immunoassay(ELISA)to detect the content of ROS,SOD and MDA in renal tissue,organize the data,and use SPSS24.0 statistical software for analysis.Result1 Effects of DDP on renal tissue morphology in mice:before treatment,the renal tubules and glomerular structure of the blank group were clear and the cells were arranged,and the exfoliated epithelial cells were visible in the renal tubular cavity of the model group,acupuncture Group and moxibustion Group,the glomerular atrophy,the cystic cavity expanded,and the blood vessel wall thickened.The results showed that the renal injury model of mice could be successfully prepared by 1 intraperitoneal injection with 0.01m L/g dose of cisplatin.2 Effects of acupuncture and moxibustion on renal tissue morphology in DDP mice:After acupuncture and moxibustion treatment,the loss of renal tubular epithelial cells in acupuncture group and moxibustion group decreased and the glomerular structure recovered compared with the model group.The results showed that acupuncture and moxibustion could improve and repair renal injury in mice caused by DDP.3 Effects of acupuncture and moxibustion on ROS content in renal tissue of DDP mice:Compared with the blank group,the content of ROS in the model group increased significantly(P<0.05),and after 5 days of acupuncture treatment,the content of ROS in acupuncture Group and moxibustion group decreased significantly compared with that of the model group(P<0.05).Compared with acupuncture group,the content of ROS in Moxibustion group was lower,and there was significant difference(P<0.05).The results showed that acupuncture and moxibustion could reduce the content of ROS,and moxibustion was better than acupuncture.4 Effect of acupuncture and moxibustion on MDA content in renal tissue of DDP mice:Compared with the blank group,the MDA content of the model group increased significantly(P<0.05),and after 5 days of acupuncture treatment,the content of MDA in acupuncture Group and moxibustion group decreased significantly compared with the model group(P<0.05).Compared with acupuncture group,the content of MDA in Moxibustion group was lower,and there was significant difference(P<0.05).The results showed that acupuncture and moxibustion could reduce the content of MDA,and moxibustion was better than acupuncture.5 Effects of acupuncture and moxibustion on SOD activity in renal tissue of DDP mice:Compared with the blank group,the SOD activity of the model group decreased significantly(P<0.05),and after 5 days of acupuncture treatment,the SOD activity of Acupuncture Group and moxibustion group increased significantly compared with the model group(P<0.05).Compared with acupuncture group,the SOD activity of Moxibustion group was higher,and there was significant difference(P<0.05).The results showed that acupuncture and moxibustion could improve SOD activity,and moxibustion was better than acupuncture.Conclusion1 Acupuncture can reduce the content of ROS and MDA in renal tissue of DDP mice,reduce the production of lipid peroxide MDA by reducing the lipid peroxidation reaction of ROS,and inhibit the damage of oxidative stress response to renal tissue.2 Acupuncture can improve the activity of SOD in renal tissue of DDP mice,inhibit and remove the expression of reactive oxygen species in renal tissue,block its lipid peroxidation reaction,reduce the formation of MDA,and achieve dynamic balance between oxidation and antioxidant.3 Compared with moxibustion,moxibustion is more favorable to repair and improve the renal tissue injury of mice caused by cisplatin by increasing the activity of SOD and reducing the content of ROS and MDA in renal tissue.
