AMPK-ACC-CPT1-based Wenyang Yiqi Recipe Promotes Myocardial Fatty Acid Oxidative Metabolism

Objective: The pathological basis of ventricular remodeling after heart failure is mainly myocardial injury.The energy supply of the injured myocardium is insufficient,and the conversion of the energy-intake substrate gradually reduces the energy supply of fatty acid oxidation and brings about adverse effects.Activation of AMPK and AMPK-ACC-CPT1 signaling promote fatty acid metabolism.Wenyang Yiqi Recipe has protective effects on myocardial injury,but its mechanism is still unclear.In this study,we established a model of H9c2 cardiomyocyte injury induced by oxygen glucose deprivation(OGD),and intervened by Wenyang Yiqi Recipe and AMPK inhibitor to explore the effect of Wenyang Yiqi Recipe on AMPK-ACC-CPT1 in injured cardiomyocytes and promote myocardial fatty acid oxidation.Methods: 1.Prepare Wenyang Yiqi Recipe for cell experiment,calculate the concentration of Wenyang Yiqi Recipe;Use CCK-8 Kit to detect the cell viability of H9c2 cells treated with different concentrations of Wenyang Yiqi Recipe,and select the appropriate and safe dose of Wenyang Yiqi Recipe;2.Establish a model of H9c2 cells injury induced by oxygen glucose deprivation.The injury was induced by different intervention durations of OGD.The morphology of injured cells was observed under light microscope,the cell viability of each group was measured by CCK-8 Kit,and the AMPK phosphorylation of injured cells was detected by Western blot.The activation level is determined to determine the duration of the OGD intervention to establish a injury model;3.Intervention of Wenyang Yiqi Recipe and AMPK inhibitor on OGD-induced injury cell model.Western blot and q PCR analysis was used to detect the phosphorylated expression of AMPK and ACC and the protein and gene expression of CPT1.4.Intervention of Wenyang Yiqi Recipe and AMPK inhibitor on OGD-induced injury cell model,microscopic observation of the effect of Wenyang Yiqi Recipe on the morphology of injured cells,and detection of cellular triglyceride(TG)and free fatty acid(FFA)content level.Results: 1.The concentration of Wenyang Yiqi Recipe for cell experiments was 0.3g/m L,and the appropriate concentration of Wenyang Yiqi Recipe for this experiment was 1.6mg/m L;2.Established a model of H9c2 cells injury induced by OGD intervention for 6 h;3.Wenyang Yiqi Recipe can significantly activate AMPK,ACC phosphorylation and CPT1 protein expression in injured H9c2 cells,and AMPK inhibitors can antagonize the activation of AMPK-ACC-CPT1 by Wenyang Yiqi Recipe.Wenyang Yiqi Recipe on injured H9c2 cells promoted CPT1 gene expression;4.Wenyang Yiqi Recipe can significantly reduce the TG level of injured H9c2 cells,but the effect of reducing FFA level was not obvious.Conclusions: 1.Wenyang Yiqi Recipe had an effect on the activation of AMPK-ACC-CPT1 signaling pathway in injured H9c2 cells;2.Wenyang Yiqi Recipe had a promoting effect on myocardial fatty acid oxidation in injured H9c2 cells;3.Activation of AMPK-ACC-CPT1 signaling pathway in H9c2 cells can activate myocardial fatty acid oxidation;4.The current experimental results can be inferred that the effect of Wenyang Yiqi Recipe on myocardial fatty acid oxidation in injured H9c2 cells was through activation of AMPK-ACC-CPT1 signaling pathway,but it cannot be fully confirmed,and further experimental studies are needed.
