Serum Inflammation Cytokine IL-1β & IL-27 Levels And The Genetic Polymorphism In Patients With The Sporadic Parkinson’s Disease

Objective:The role of neuroinflammation in PD and relations between the following four SNPs and PD susceptibility shall be further explored by observing serum inflammation cytokine IL-1β & IL-27 levels in patients with Sporadic Parkinson Disease among Han people,and analyzing SNPs including rs16944(IL-1β),rs153109,rs17855750,and rs181206(IL-27),which shall provide theoretical foundation for seeking for possible PD biomarkers and new treatments.Methods:We used the modified Hoehn and Yahr(H and Y)scale to evaluate severity of motor symptoms of 52 SPD patients.ELISA method was used to measure serum inflammation cytokine IL-1β and IL-27 levels in the PD group and the control group,and observed the differences in IL-1β and IL-27 levels between PD patients with different clinical phenotypes such as age,gender,course of disease,disease severity,and initial symptoms,etc.Multiplex PCR technique was used to test the rs16944 of IL-1β,and rs153109,rs17855750,rs181206 of IL-27,and the differences in genetic polymorphism from the initial symptoms of PD patients.Results:(1)Compared with the control group,serum IL-1β level in patients from the PD group was significantly higher,while IL-27 level was significantly lower,which had statistical significance(P<0.001).(2)According to the different courses of the disease,there were differences in serum IL-1β and IL-27 levels of PD patient(P<0.001),among which the IL-1β levels and IL-27 levels of patients with the disease for 5-10 years or for more than 10 years were higher than that of patients suffering from the disease for less than 5 years,and the difference had statistical significance(P < 0.05);while there was no difference in IL-1β and IL-27 levels between patients having the disease for 5-10 years and those suffering from the disease for more than 10 years(P>0.05).(3)Serum IL-1β and IL-27 levels in PD patients were positively related to dyskinesia severity(P<0.05).(4)There was no significant difference in serum IL-1β and IL-27 levels between PD patients with different gender,ages and motor phenotypes(P>0.05).(5)There was no difference in the serum protein levels with different genotypes of rs16944 of IL-1β(P>0.05);there was also no difference in the serum IL-27 levels with different genotypes of rs153109,rs181206 and rs17855750(P > 0.05).(6)There was difference being found in genotypic distribution frequency and allele distribution frequency of rs16944 of IL-1β between PD group and the normal control group(P<0.05),but there was no significant difference in genotypic distribution frequency and allele distribution frequency of rs16944 of IL-1β between PD patients with different motor phenotypes(P > 0.05).(7)There was no difference in genotypic distribution frequency and allele distribution frequency of rs153109,rs181206 and rs17855750 in terms of IL-27 between PD group and the normal control group,and between PD patients with different motor phenotypes(P>0.05).Conclusion:(1)Inflammatory cytokines IL-1β and IL-27 may be involved in the generation and development of PD,and serum IL-1β and IL-27 levels are related to motor symptoms severity and the course of PD.(2)The serum levels and genetic polymorphism of IL-1β and IL-27 may not be correlated to PD motor phenotypic tremor.(3)Polymorphism of rs16944(IL-1β)is correlated to PD,and IL-1β may be a susceptibility gene of SPD;polymorphism of rs153109,rs181206 and rs17855750(IL-27)may not be related to the generation of SPD.
