Effect Of Mig-7 On Glioma Migration,Invasion,Proliferation And Vasculogenic Mimicry And Its Mechanism | | Posted on:2021-02-11 | Degree:Master | Type:Thesis | | Country:China | Candidate:Z G Pan | Full Text:PDF | | GTID:2504306128970699 | Subject:Outside of the surgery (God) | | Abstract/Summary: | PDF Full Text Request | | Objective: To investigate the regulatory role of MAPK pathway in the process of mig-7promoting glioma migration,invasion and proliferation and the formation of vasculogenic mimicry.Methods: Collect clinical glioma specimens(60 cases),respectively using Western blot and CD34-double staining PAS was detected in protein of Mig-7 and the expression of VM.To cultivate normal glial cell lines HA1800 and glioma cell lines: H4 SWO38 U87 MG,detection of four strains of Mig-7 protein and mRNA expression in cells.Small interference RNA transfection Mig-7 U87 MG cell lines,invalid sh RNA as negative control.mRNA and protein expression of vm-related genes(mmp-2,mmp-9,mmp-14,LAMC2,Eph A2,ve-cadherin)in blank,sh-NC and sh-Mig-7 groups were detected.The mig-7-pc-DNA up-regulated by mig-7 was transferred to glioma cell line U87 MG by lentivirus infection system,invalid nc-pcdna was used as a negative control.Six groups in the blank group oe-NC group,oe-mig-7+SP600125(JNK inhibitor)group,oe-mig-7+SCH772984(ERK inhibitor),oe-mig-7 + SB202190(p38 inhibitor)were tested for migration and invasion ability and proliferation ability.Results:The expression of VM mig-7 protein in glioma tissues of 60 cases increased with the increase of WHO pathological grade,and the two were positively correlated.mRNA and protein levels of glioma cell line mig-7 were significantly up-regulated compared with normal glial cell line HA1800(P<0.05).mRNA and protein expression of vm-related genes(mmp-2mmp-9 mmp-14 LAMC2 Eph A2 ve-cadherin)were significantly decreased in the sh-mig-7group compared with the blank and sh-NC group(P < 0.05).Detection of migration and invasion ability detection of proliferation ability test results showed that after overexpression of mig-7,the cell migration,invasion and proliferation VM formation ability was significantly enhanced(P < 0.05).Three MAPK path-related inhibitors,SP600125,SCH772984 and SB202190,can partially inhibit this process(P<0.05).Conclusion:Mig-7 mediates mapk-related pathways to affect VM formation,proliferation,migration and invasion of glioma. | | Keywords/Search Tags: | Glioma, migration-induced gene-7, migration, invasion, proliferation, vasculogenic mimicry, MAPK | PDF Full Text Request | Related items |
