| ObjectiveIn this study,intraperitoneal injection of doxorubicin(doxorubicin)was used to establish a rat model of heart failure,and to explore the significance of β3-AR,PI3 K,and pAkt protein expression in the myocardium of heart failure rats.Method14 clean Wistar male rats(<6 weeks)were randomly divided into 9 heart failure groups and 5 control groups.The rats in the heart failure group were given intraperitoneal injection of doxorubicin at a dose of 1 mg / kg;the rats in the control group were intraperitoneally injected with an equal amount of normal saline twice a week for 8 weeks.General conditions,initial body weight,final body weight,exercise tolerance,heart weight,left ventricular muscle thickness,ascites volume,and the NT-pro BNP value and free triiodothyronine(FT3)value in the blood of rats were measured.Correlation analysis of protein expression of β3-AR,PI3 K and pAkt in rat myocardium.ResultThe model of adriamycin-induced heart failure in rats was successfully constructed.The rats in the heart failure group gradually reduced food intake,slow or even weight gain,hair loss,and decreased vitality.The initial weight of the heart failure group was similar to that of the control group(100.0(97.0,102.0)vs 102.0(96.0,104.0)g,P =0.639);the final body weight of the heart failure group was significantly reduced compared with the control group(284.0(260.3,284.1)vs 357.5(342.6,368.1)g,P =0.003);the time of staying at the A terminal of the heart failure group was significantly shorter than that of the control group(19(17,27)vs 36(33,46)seconds,P = 0.003);the heart weight of the heart failure group was higher than that of the control group Rats relieved(0.8(0.8,1.0)vs 1.2(1.05,1.3)g,P = 0.010);cardiac mass index of rats in heart failure group and control group(0.0043(0.0040,0.0062)vs 0.0049(0.0039,0.0052),P = 0.639);The maximum thickness of left ventricular muscle in the heart failure group was lower than that in the control group(0.24(0.19,0.25)vs 0.43(0.40,0.46)cm,P = 0.003);the heart failure group was larger The amount of ascites in rats was significantly higher than that in the control group(41(32,59)vs 2(2,5)ml,P =0.003).The NT-pro BNP in the blood of the heart failure group was significantly higher than that of the control group(the OD value of NT-pro BNP in the heart failure group and the control group(0.678(0.605,0.720)vs 0.620(0.618,0.650),P = 0.03);In the heart failure group,non-coagulated pink ascites was seen in the abdominal cavity.The content of FT3 in the blood of the heart failure group rats was similar to that of the control group(heart failure and control group FT3 OD value 0.360(0.327,0.373)vs0.349(0.316,0.362),P = 0.53).Western blot results showed that the expression ofβ3-AR protein in the heart failure group was significantly higher than that in the control group(gray value: 0.884(0.818,1.048)vs 0.481(0.408,0.514),P = 0.009),heart failure The expression of PI3 K protein in myocardium of group rats was significantly higher than that of control group(gray value: 1.055(0.911,1.117)vs 0.426(0.348,0.488),P =0.008).The expression of pAkt protein in the heart failure group was significantly higher than that in the control group(gray value: 0.873(0.801,1.258)vs 0.242(0.182,0.382),P = 0.008).There is a linear positive correlation between β3-AR and PI3 K,and there is a linear correlation between β3-AR and pAkt.Conclusion1.Wistar rats were intraperitoneally injected with 1 mg / kg doxorubicin hydrochloride to successfully establish a rat model of heart failure.2.No increase in FT3 value,suggesting that doxorubicin does not damage myocardial cells through oxidative stress.3.The expression of β3-AR protein in the heart failure group was significantly higher than that in the control group,suggesting that β3-AR protein was involved in the process of worsening heart failure.4.PI3K,pAkt and β3-AR have a linear positive correlation,suggesting that there may be an isotropic relationship between β3-AR and PI3 K / Akt pathway in myocardial cells of heart failure rats. |
