Background: Long-term Western eating habits are closely related to the occurrence and development of metabolic diseases,and Gut microbial disorders play a key role in this,Modern studies have shown that Ligustrum robustum(Roxb.)Blume can regulate body metabolism,However,the effects of it on normal diet and western diet mice gut microbiota have not been reported.Methods: The experiment was divided into normal diet group(ND),western diet group(WD),normal diet +(18g / kg)Ligustrum robustum(Roxb.)Blume high-dose group(NKDT),Western diet + 2% Ligustrum robustum(Roxb.)Blume free drinking group(WFKDT),Western diet +(6g / kg)Ligustrum robustum(Roxb.)Blume low-dose group(WLKDT),Western diet +(12g / kg)Ligustrum robustum(Roxb.)Blume medium-dose group(WMKDT),Western diet +(18g / kg)Ligustrum robustum(Roxb.)Blume high Dose group(WHKDT),According to the experimental plan,the model was built and administered.During the experiment,the body weight was measured once a week.At the end of the 10 th week,the contents of serum and colon were taken.Detecting the content of serum TG,TC,LDL-C,HDL-C,Glu,ALT,AST;Detecting microbial diversity in colon contents(only ND,WD,NKDT,WFKDT,WHKDT detection)by16S r DNA Amplicon Sequencing.Results: 1.The body weight at 10 th week,serum Glu,ALT,AST,TG,TC,LDL-C of NKDT mice were not significantly different from those of ND mice;Compared with ND mice,the abundance of Alloprevotella increased(P<0.05),the abundance of Alisipes and Lachnospiraceae all decreased(P<0.05)in the gut microbiota of NKDT mice.2.The body weight at 10 th week,serum Glu,ALT,AST,TG,TC,and LDL-C of WD mice were higher than those of ND mice(P<0.001);Compared with ND mice,the abundance of Firmicutes,Erysipelotrichia,Erysipelotrichales,Erysipelotrichaceae,Allobaculum,Proteobacteria,Gammaproteobacteria,Burkholderiaceae,Parasutterella,Bacteroidaceae and Bacteroides all increased(p<0.001),the abundance of Bacteroidetes,Rikenellaceae and Alistopes all reduced(p<0.001),The abundance of Ruminococcaceae decreased(p<0.05)in the gut microbiota of WD mice.3.Compared with WD mice,the body weight decreased at 10 th week(P<0.001),Glu,ALT,and AST all decreased(P<0.01),and TG decreased(P<0.05)in the WFKDT mice;the abundance of Ruminococcaceae,Rikenellaceae and Alisipes increases(P<0.01),the abundance of Bacilli,Lactobacillales,and Lactobacillus all decrease(P<0.05)in the gut microbiota of WFKDT mice.4.Compared with WD mice,the body weight decreased at 10 th week(P<0.001),ALT,AST,and TG all decreased(P<0.01)in the WLKDT mice.5.Compared with WD mice,the body weight decreased at 10 th week(P<0.01),AST decreased(P<0.05),TC increased(P<0.01),LDL-C increased(P<0.05)in the WMKDT mice.6.Compared with WD mice,the body weight(P<0.01),AST decreased(P<0.05),TC increased(P<0.01),LDL-C increased(P<0.05)in the WHKDT mice;The abundance of Verrucomicrobia,Akkermansia,Deltaproteobacteria,Desulfovibrionaceae and Lachnoclostridlum all increased(P<0.01);The abundance of Erysipelotrichia,Erysipelotrichaceae all reduced(P<0.001)in the gut microbiota of WHKDT mice.Conclusion: This study shows that a moderate amount(6g/kg or 2%)of Ligustrum robustum(Roxb.)Blume can improve the transaminase,triglyceride abnormalities and related gut microbiota disorders caused by Western diet in mice,but medium and high doses(>6g/kg)Ligustrum robustum(Roxb.)Blume will aggravate the abnormalities of total cholesterol,low-density lipoprotein cholesterol and related gut microbiota disorder caused by Western diet.The mechanism of the effect of different dosages of Ligustrum robustum(Roxb.)Blume on the gut microbiota of western diet mice needs further research.
