Objective: To investigate the effect of celecoxib on the formation of Staphylococcus aureus biofilm on the surface of surgical implants in vitro.Methods: We retrieved orthopedic infections from Wuhan Union Medical College Hospital during 2018 and analyzed the bacterial distribution.Tissue culture plate method was used to measure the biofilm formation ability of four S.aureus strains including methicillin-resistant S.aureus.Spinal surgical implants were processed into titanium rods for quantitative experiments,and titanium sheets were subjected to morphological experiments,maintaining the same surface area and roughness.The minimum inhibitory concentration of non-steroidal anti-inflammatory drugs was obtained by the broth dilution method.Quantitative detection of biofilms on titanium rods was performed by enzyme treatment and conventional plate counting method.Scanning electron microscope was used to observe the morphological distribution of bacteria on titanium plates.The morphological method used scanning electron microscope to observe the distribution of bacteria on titanium plate.Quantitative and morphological methods were used to explore the effects of celecoxib on S.aureus adhesion to implants,biofilm formation,and mature biofilms.Results: The orthopedic infection in Union Hospital was mainly Staphylococcus aureus,accounting for 26.3%,of which methicillin-resistant S.aureus was 32.9%.The selected strains in the experiment had strong-medium biofilm formation ability.Celecoxib and other non-steroidal anti-inflammatory drugs,such as aspirin,ibuprofen,indomethacin,and diclofenac sodium,all had the effect of inhibiting bacterial reproduction and antibacterial biofilms.The effects of these drugs were dose-dependent.The minimum inhibitory concentration of celecoxib was 25-50 ug/ml.12.5ug/ml celecoxib reduced the amount of bacteria by 10-15 times in the adhesion experiment,reduced the amount of bacteria in the new biofilm by 80-200 times,and reduced the amount of bacteria in the mature biofilm by 14-30 times.Scanning electron microscopy results showed that 12.5 ug/ml celecoxib inhibited the formation of bacterial biofilms,and 25 ug/ml celecoxib could clear the mature biofilms formed by bacteria on titanium rods.Conclusion: Our recent research found that celecoxib can resist biofilm infection in vitro,and it may be achieved by inhibiting bacterial adhesion and biofilm formation and destroying mature biofilms.Therefore,Non-steroidal anti-inflammatory drugs are expected to become adjuvant drugs for the treatment of plant-related infections in the spine.
