| Triple negative breast cancer(TNBC)is characterized by non-expression of ER,PR,and HER2.TNBC is highly invasive and has poor clinical prognosis.LncRNAs play an important role in the process of TNBC.In-depth study of the biological characteristics of lncRNAs and its role in invasive metastasis are expected to provide new targets for early diagnosis and prognostic monitoring of TNBC.By downloading the sequencing data and clinical information of breast cancer in the TCGA database,we selected the TNBC and cancer adjacent data for differential expression analysis and cox survival analysis.The results showed that the expression levels of 175 genes(including 40 LncRNAs)were related with the survival of patients with TNBC,and the lncRNA HAGLROS gene had the highest HR value(HR = 4.75,95CI%159-14.24,p = 0.005).The results of biological experiments showed that HAGLROS was highly expressed in TNBC cells,and knockdown of HAGLROS could significantly inhibit the proliferation and migration ability of MDA-MB-231 cells(TNBC cells),suggesting that HAGLROS may play an important role in the development of TNBC.Through co-expression and online database of lncRNA and mi RNA for bioinformatics analysis,we predicted that HAGLROS may regulate 8 m RNA genes(LRRC8D,HOXD13,TMPRSS4,HRAS,HOXD8,LRP4,HOXD9,TFF3);in addition,HAGLROS may target7 mi RNAs(hsa-mi R-326,hsa-mi R-330-5p,hsa-mi R-1297,hsa-mi R-4750,hsa-mi R-6727,hsa-mi R-6749,has-mi R-6799)to regulate 23 m RNA genes(PKNOX2,SIDT1,PRDM16,HOXD13,NGFR,TMEM179,DMBX1,KLHDC7 A,KCNQ4,ADAMTS15,KCNJ12,ATP2B3,PAX5,DCLK1,CNR1,MYT1,MAN1C1,RXRG,FOS,SLC20A1,CNTNAP2,SALL3,SCN9A),KEGG analysis showed that the target m RNAs were mainly involved in 5 signaling pathways.According to the predicted ce RNA network(1 lncRNA,7 mi RNA,30 m RNA),the preliminary functional verification was carried out,and the results showed that the expression levels of hsa-mi R-6799-3p and hsa-mi R-330-5p were up-regulated after knocking down HAGLROS,while the expression levels of hsa-mi R-4750-5p,hsa-mi R-6749-5p,hsa-mi R-6799-3p and hsa-mi R-330-3p were down-regulated after overexpression of HAGLROS,it is suggested that HAGLROS may affect the proliferation and metastasis of TNBC cells by sponging adsorption hsa-mi R-6799-3p.In summary,using the TCGA database,we revealled that HAGLROS was strongly related to the prognosis of TNBC,HAGLROS may affect the proliferation,and metastasis of TNBC cells through hsa-mi R-6799-3p sponge activity,which could provide new ideas for the treatment of TNBC. |
