Diterpenoids are terpenoids containing four isoprene units,with a total of 20 carbon atoms.They are a class of natural products with complex and diverse structures and important biological activities.They are widely distributed in higher plants,fungi,insects and the marine organism.In higher plants,diterpene compounds are mainly found in some gymnosperms and Compositae,Labiatae,Euphorbiaceae,Ranunculaceae,Annonaceae,Zingiberaceae,Rutaceae and so on.Diterpene compounds are of various types and rich in structure types,including kaurane-type,labdane-type,clerodane-type,abietane-type and pimarane-type,etc.,and have various biological activities and applications.Modern pharmacological activity studies have shown that diterpene compounds have good antipyretic,anti-inflammatory,antibacterial,anti-viral,anti-cardiovascular disease and anti-tumor,cytotoxic,antimalarial,anti-oxidant,anti-bacterial,and enzyme inhibitory activities.Gochnatia is one of the two rare woody plants in the Compositae,and Gochnatia decora(Kurz)A.L.Cabrera is one of them.As a folk medicine with a long history,It is mainly distributed in southern Yunnan in China,and is used to treat cough with lung heat,asthma,stab wounds,and gunshot wounds.In the early stage,the research team isolated a series of enantio-carpelane-type diterpenoids from the bark and branches of G.decora(Kurz)A.L.Cabrera,and found that most of them can inhibit the production of NO by LPS-induced mouse macrophage RAW 264.7.In order to further study whether the G.decora(Kurz)A.L.Cabrera and these compounds have anti-inflammatory activity,this paper continues to use the LPS-induced mouse macrophage RAW 264.7 model to research 19 diterpenoids compounds from G.decora(Kurz)A.L.Cabrera in vitro for antiinflammatory activity.After cytotoxicity test,NO inhibition test,ELISA test,Westren Blot test,14 diterpenoids compounds were found to inhibit NO production and reduce inflammatory factors(TNF-α,IL-1β,IL-6,IL-17)expression,down-regulate protein(COX-2,i NOS)expression level and inhibit the phosphorylation of NF-κB p65.The above results suggest that the diterpenoids in white chrysanthemum have potential anti-inflammatory activity,which provides a scientific basis for exploring the active substance basis in white chrysanthemum.Secondly,the LPS-induced mouse acute pneumonia model was used to study the anti-inflammatory activity of crude extracts from the branches and leaves of G.decora(Kurz)A.L.Cabrera.Mice were gavaged with crude methanol extracts from the branches and leaves of G.decora(Kurz)A.L.Cabrera for one week,and infected with LPS for 48 hours.Lung tissues were collected,and the morphology of the lung tissues of each group was observed by HE staining,lung wet dry weight ratio,ELISA and other experimental methods.The changes of chemistry and the content of inflammatory factors were used to evaluate the effect of crude methanol extracts of A.chrysanthemum on the LPS-induced acute pneumonia in mice.The results found that: compared with the model group,the crude methanol extract of the branches and leaves of G.decora(Kurz)A.L.Cabrera can effectively improve the morbidity of acute pneumonia mice and reduce the expression level of inflammatory factors in lung tissue.The folk medicine has a long history of use and definite curative effect.Starting from its traditional efficacy,using modern chemistry and pharmacology research methods to elucidate its substance basis and mechanism of action is of great significance to the scientific development and utilization of folk medicine.In this paper,the anti-inflammatory activity of diterpene monomer compounds and their extracts in G.decora(Kurz)A.L.Cabrera is evaluated by using a model of inflammation in vivo and in vitro,which provides an early basis for the research on the pharmacodynamic substances and mechanism of action of G.decora(Kurz)A.L.Cabrera.
