| This thesis consists of four parts.The first part summarizes the current research status of influenza and anti-influenza drugs and the research progress of Sophora and Laggera pterodonta.The chemical constituents and biological activities of Sophora viciifolia in the second chapter are discussed in detail.The chemical constituents and anti-influenza activity of Laggera pterodonta in the third chapter are discussed in detail.The fourth part summarizes and discusses the work in this paper.Influenza is caused by influenza virus,which has the characteristics of strong infectivity,rapid epidemic and high incidence,Influenza pandemics have a serious impact on the safety of people’s lives and property and social stability around the world,and traditional anti-influenza drugs have varying degrees of drug resistance and toxic side effects.Therefore,it is necessary to seek treatment methods and drugs with good curative effect and low toxicity from natural products.Traditional Chinese medicine has a long history and outstanding advantages in the treatment of influenza.Both Sophora plants and Laggera pterodonta have anti-influenza active ingredients.This paper reviews the research progress of Sophora and Laggera pterodonta,which provides a research basis for finding more effective anti-influenza active ingredients.S.viciifolia is a plant of the genus Sophora,which is distributed in many provinces in China.In most areas of Yunnan,there is the custom of eating flowers of S.viciifolia,which has the effects of clearing away heat and detoxification and reducing swelling.We studied the chemical constituents of the flowers of S.viciifolia,and isolated and identified 30 quinolizidine alkaloids from its total alkaloids,including 22 matrine-type alkaloids and 6 sparteine-type alkaloids and 2 cytisine-type alkaloids.Six new compounds were identified by means of 1D-,2D-NMR,and HR-ESI-MS.They were named sophovicine A(1),8α-hydroxysophocarpine(2),13β-hydroxyoxymatrine(3),9α-acetoxymatrine(4),oxysophoramine(5),14β-hydroxylupanine(6),where compound 1 is a novel Matrine-type derivative with a C20N7 skeleton.All the isolated compounds were screened for anti-H1N1 influenza virus and anti-inflammatory activity in vitro,and the results showed that they did not show significant anti-influenza and anti-inflammatory activity.L.pterodonta is a plant of the genus Laggera,which is mainly distributed in Yunnan and Sichuan provinces.It is often used to treat colds,pharyngitis,bronchitis and other diseases.At present,there are a variety of L.pterodonta preparations or Chinese patent medicines with L.pterodonta as the main component.The chemical constituents in L.pterodonta are mainly flavonoids and sesquiterpenes.We studied the chemical constituents of L.pterodonta,and isolated and identified 10 compounds,including 1 new eosane-type sesquiterpene,7 flavonoids and 2 lignans.Petroleum ether phase,ethyl acetate phase,different polar phases of ethyl acetate phase,and n-butanol phase obtained by extraction of crude extract of L.pterodonta and all the separated monomer compounds from L.pterodonta were screened against H1N1 influenza virus in vitro,and the results showed that none of them showed obvious anti-influenza activity.Finally,summarize and discuss the work done in this paper.Through extraction,separation and structural identification of S.viciifolia and L.pterodonta,40 compounds were identified,including 7 new compounds.The structural types of these compounds include quinolizidine alkaloids,flavonoids and sesquiterpenes.In the key research on S.viciifolia,several new quinolizidine alkaloids were found,especially a novel matrine-type derivative with a C20N7 skeleton.These findings not only deepened the understanding of the chemical composition of S.viciifolia,but also enriched the skeleton type of quinolizidine alkaloids.The further structural modification and modification of such alkaloids are of great significance for obtaining better activity and developing innovative drugs. |
