ObjectiveWith the economic growth and the improvement of living standards,the prevalence of obesity is increasing.It is called "death Quartet" together with hypertension,hyperlipidemia and hyperglycemia.In order to deal with the worldwide medical and social problems caused by obesity,more efficient and safe treatment methods are urgently needed.Smiax glabra Roxb.is the dried rhizome of Rhizoma Smilacis Glabrae,which belongs to Liliaceae.As root and stem medicine,it contains a lot of polysaccharide.However,polysaccharide is a natural product with a wide range of sources.It has the advantages of small side effects,no residue,and is not easy to produce drug resistance.Studies have shown that polysaccharide from Smiax glabra Roxb.has anti-inflammatory and immunomodulatory effects.However,there are no reports about it in reducing weight and fat at home and abroad.Therefore,further study on polysaccharide from Smiax glabra Roxb.can provide theoretical basis and scientific experimental basis for further development and utilization of Smiax glabra Roxb.and prevention and treatment of obesity.In this study,the model organism of caenorhabditis elegans was used to study the anti fat and anti-oxidation activities of polysaccharide from Smiax glabra Roxb.Methods1.Taking caenorhabditis elegans bristol(N2)as a living model,the effects of different concentrations of polysaccharide from Smiax glabra Roxb.on its fat metabolism were investigated:the effects of pachyman on fat accumulation of caenorhabditis elegans bristol(N2)were evaluated by oil red 0 staining and Sudan black staining;ROS level of caenorhabditis elegans bristol(N2)was measured by reactive oxygen fluorescence probe.The activities of TG and SOD,the content of MDA and the expression of genes related to fat metabolism were studied in order to explore the mechanism of weight loss and lipid reduction of polysaccharide from Smiax glabra Roxb.2.The model was a knockout mutant of caenorhabditis elegans,NHR-80(BX165),to determine the effect of pachyman on the TG content of caenorhabditis elegans,and further clarify the possible mechanism.Results1.The results of oil red 0 staining showed that,compared with the blank group,the coloration range of caenorhabditis elegans bristol(N2)in different concentrations of polysaccharide from Smiax glabra Roxb.and orlistat group was small,the color was light,and the accumulation of lipid droplets was significantly less.Compared with the orlistat group,the staining of the subcutaneous tissue of caenorhabditis elegans bristol(N2)was significantly reduced,and the lipid droplets were mainly concentrated in the intestinal tract,and the staining was shallower,and the lipid droplets were less concentrated.The results of Sudan black staining showed that,compared with the blank group,the color in the body of caenorhabditis elegans bristol(N2)in the groups treated with different concentrations of polysaccharide from Smiax glabra Roxb.and orlistat was lighter,and the lipid droplets were less.But the difference was not obvious compared with oil red 0 staining.Compared with the blank group,after administration of different concentrations of polysaccharide from Smiax glabra Roxb.group and orlistat group,the content of TG in the body of caenorhabditis elegans bristol(N2)decreased(P’<0.01).2.Compared with the blank group,after administration of different concentrations of polysaccharide from Smiax glabra Roxb.group and orlistat group,the fluorescence intensity of polysaccharide from Smiax glabra Roxb.and orlistat group in caenorhabditis elegans bristol(N2)decreased significantly.With the increase of drug concentration,the fluorescence intensity decreased;Compared with the blank group,after administration of different concentrations of polysaccharide from Smiax glabra Roxb.group and orlistat group,the content of MDA in the body of caenorhabditis elegans bristol(N2)decreased(P’<0.01),SOD content increased(P’<0.01 or P’<0.05).3.Compared with the blank group,after administration of different concentrations of polysaccharide from Smiax glabra Roxb.group and orlistat group,the results of RT-qPCR showed that the expression of FAT-2,NHR-80 and SBP-1 mRNA in the caenorhabditis elegans bristol(N2)was significantly decreased,and the results were statistically different(P’<0.01 or P’<0.05).4.The effect of polysaccharide from Smiax glabra Roxb.with different concentrations on the TG content in the knockout mutant of caenorhabditis elegans NHR-80(BX165)was determined,the results showed that there was no significant difference in TG content between the groups.(P’>0.05).ConclusionPolysaccharide from Smiax glabra Roxb.can significantly reduce the accumulation of lipid droplets and TG in the body of caenorhabditis elegans bristol(N2),suggesting that polysaccharide from Smiax glabra Roxb.can reduce the accumulation of lipid in the body of caenorhabditis elegans bristol(N2).In addition,polysaccharide from Smiax glabra Roxb.can significantly reduce the content of ROS and MDA,and increase the content of SOD,suggesting that it can effectively alleviate the lipid peroxidation level of caenorhabditis elegans bristol(N2).Meanwhile,polysaccharide from Smiax glabra Roxb.inhibited the gene expression of FAT-2,NHR-80 and SBP-1 in caenorhabditis elegans bristol(N2).After the NHR-80 gene was knocked out,the TG content of caenorhabditis elegans could not be reduced.It is suggested that polysaccharide from Smiax glabra Roxb.can reduce fat accumulation by inhibiting gene expression of FAT-2,NHR-80 and SBP-1,and its mechanism may be more related to NHR-80.All in all,polysaccharide from Smiax glabra Roxb.has obvious activity of reducing weight and fat.Its regulation mechanism may be mainly through inhibiting gene expression of FAT-2,NHR-80,SBP-1,desaturating saturated fatty acids,inhibiting lipid synthesis,enhancing metabolic efficiency,effectively alleviating lipid peroxidation level and reducing fat accumulation in Caenorhabditis elegans,so as to achieve the goal of reducing weight and fat.
