Objective:Through the investigation and statistical analysis of genetic polymorphism of 20 autosomal STR loci in Chinese Korean population in Yanbian area,the allele and genotype distribution frequencies and related population genetic parameters were investigated to observe the applicability of 20 STR loci in forensic identification in Chinese Korean population.And by comparing with other groups at home and abroad,the ethnic genetic characteristics of Chinese Korean population were determined,providing basic data for the establishment of genetic database of Korean population in China.Methods:The oral swabs of 200 Korean unrelated individuals in Yanbian area were collected,and the DNA of the samples was extracted by Chelex-100 method.According to the instructions of the PowerPlex21 System kit,the DNA samples obtained were subjected to compound amplificat ion.The amplified products were subjected to capil l ary electrophoresis detection using the ABI PRISM 3100 automatic genetic analyzer,and the data was analyzed using GeneMapper ID 3.2 genetic analysis software and WEN ILS 500 internal standard.Handling and typing.Use the Modified-Powerstates standard version to perform statistics on forensic genetic parameters such as allele and genotype frequency,heterozygosity,personal recognition,genetic diversity,and non-father exclusion rate and Hardy-Weinberg equilibrium test.At the same time,the use of POPTREE2 software was conducted on 13 groups including Chinese Korean and Korean,Japanese and domestic Han,Miao(2 regions),Tujia,Kazakh,Hui,Mongolian,Gelao,Hani,Dai,Zhuang,etc.Contrast analysis of genetic polymorphism,calculate the genetic distance between groups,and draw phylogenetic tree and UPGMA cluster analysis according to the research model designed by Neighbour-Joining(NJ)method.Results:The results of genetic polymorphism survey of 200 Chinese Korean populations in Yanbian area showed that a total of 220 alleles were detected in 20 autosomal STR loci,with allele frequencies distributed between 0.003 and 0.515;a total of 675 genotypes were detected,with frequencies distributed between 0.005 and 0.320;the total amount of polymorphic information(PIC)was distributed between 0.600 and 0.910,heterozygosity(H)was distributed between 0.640 and 0.950(mean heterozygosity was 0.691),personal identification power(DP)was distributed between 0.809 and 0.984,cumulative personal identification power(TDP)was greater than 0.999999999,non-paternal exclusion rate(PE)was distributed between 0.342 and 0.898,and cumulative non-paternal exclusion rate(CPE)was greater than 0.999999999.The results of genetic polymorphism comparison between Chinese Korean in Yanbian area and other 13 ethnic groups showed that the genetic distance between Korean population of the same ethnicity was the closest(0.006),and the genetic distance between Zhuang nationality was the farthest(0.119).The phylogenetic tree shows the results.Yanbian Koreans,Koreans and Japanese are grouped together,Guizhou and Yunnan Miaos are grouped together,Yunnan Zhuang,Yunnan Hani and Yunnan Dais are grouped together,Guizhou Gelao and Hunan Tujias And the Han people in Henan are classified into one group,the Hui people in Ningxia,Mongolians in Inner Mongolia and Kazakhs in Xinjiang are classified into one group,which conforms to the rules of group differentiation between different ethnic groups and between different regions.Conclusion:The 20 autosomal STR loci alleles and genotypes of the PowerPlex21 System are evenly distributed,with high personal recognition and non-father exclusion rates,and are suitable for Chinese Korean ethnic group forensic identification and genetic research.Yanbian Koreans and other groups have different degrees of difference.The results of this study not only provide basic data for the establishment of the Korean genetic database,but also clarify the necessity of establishing a population database.
