| Objective:To investigate the role of cryptotanshinone(CTS)in ovalbumin(OVA)-induced asthmatic mouse model,and explore its possible mechanism.Methods:Fifty female BALB/c mice were randomly divided into five groups:Control group(normal saline group),OVA group(OVA-induced asthma model group),and CTS 50 group(cryptosatanone low-dose group,administered at 50 mg/kg),CTS 100 group(cryptosatanone high-dose group,administered dose of 100 mg/kg),DEX group(dexamethasone,positive control group,administered at 10 mg/kg).Asthma mouse models were established by OVA sensitization and challenge,and the mice were administered intraperitoneally with 50 mg/kg,100 mg/kg CTS and 10 mg/kg dexamethasone in CTS 50,CTS 100 and DEX groups.Mice were sacrificed 24 hours after the last challenge,and bronchialveolar lavage fluid(BALF)and lung tissues of the mice were taken from each group.The levels of Th2 cytokines IL-4,IL-5,IL-13 and Th1 cytokines IFN-γ in BALF supernatants were measured,and the number of inflammatory cells in BALF precipitation was determined.After homogenizing the right lung tissues,the expression levels of Th1 cytokines IFN-y and Th2 cytokines IL-4,IL-5 and IL-13 were determined.The left lung tissues were embedded in sections and stained with HE,PAS and NF-κB p65,p-p38 MAPK immunohistochemical staining.Western blot was used to observe the effects of CTS on the levels of NF-κB p65,p-IκBα and p-p38 MAPK in lung tissue of OVA-induced mice.Results:Cryptotanshinone can significantly inhibit the total number of inflammations and various types of inflammatory cells in BALF precipitation in mouse models of OVA-induced asthma,inhibit the Th2 cytokines IL-4,IL-5,IL-13 and increase Th1 Cytokine IFN-y levels in BALF supernatant and lung tissue.HE staining results show that cryptotanshinone can significantly inhibit the infiltration of inflammatory cells around the airway,and PAS staining results show that cryptotanshinone can inhibit airway mucus secretion.Immunohistochemical results showed that cryptotanshinone can inhibit the positive expression of NF-κB p65 and p-p38 MAPK in the lung tissue of the mouse model of OVA-induced asthma.Western blot results showed that cryptotanshinone can inhibit the activation of NF-κB p65 in the lung tissue of the mouse model of asthma induced by OVA and inhibit the phosphorylation of IκBα.Conclusion:CTS can inhibit the inflammatory response in OVA-induced asthma model mice,and its mechanism may be related to the inhibition of the p3 8 MAPK/NF-κB signaling pathway. |
