ObjectiveIn order to investigate the role of β-1,3-galactosyltransferase 2(B3galt2)gene in blood-brain barrier,we compared the main structural components that are tight junction protein,cerebrovascular endothelial cells and pericytes of the blood-brain barrier in the hippocampus of wild-type mice,B3galt2 single gene knockout and B3galt2 double gene knockout mice.The study will provide a theoretical basis for studying the role of B3galt2 and other glycosyltransferases in the blood-brain barrier in the future.MethodsThe β-1,3-galactosyltransferase 2 gene knockout heterozygous mice were bred,and tail clipping was used to identify the genotyping on postnatal day 21 mice by PCR method of genomic DNA.We divided experimental mice into six groups randomly.To minimize errors between groups,adult(3 to 6-month)wild-type,heterozygous,and homozygous mice from the same litter were used for each of the experimental groups.Then randomly selected three groups of mice for immunohistochemical staining,and the other three groups for western blotting,we investigated the changes of endothelial cells,tight junctions,and pericytes in the hippocampus.Results1.B3Galt2 gene knockout significantly reduced the expression of tight junction protein in the hippocampus of adult mice,and statistical results show that P <0.05 is statistically significant.2.B3Galt2 gene knockout significantly reduced the expression level of endothelial cells in hippocampus of adult mice,and statistical results show that P <0.05 is statistically significant.3.B3Galt2 gene knockout significantly reduced the expression of pericytes in the hippocampus of adult mice,and statistical results show that P <0.05 is statistically significant.4.B3Galt2 is colocalized with tight junction and pericytes.Conclusions1.B3Galt2 gene knockout affects the major compoments of the blood-brain barrier in the hippocampus of adult mice.2.The B3Galt2 gene may be expressed in tight junctions and pericytes in the hippocampus of adult mice.
