ObjectiveTo investigate the protective effects of different doses of Unfractionated Heparin(UFH)on sepsis-induced acute lung injury(ALI)in rats.MethodsBlood gas analysis was performed to observe the change of pressure of oxygen(Pa O2).The levels of TNF-α,IL-1β and IL-6 were determined by enzyme-linked immunosorbent assay(ELISA).The expression of Rho A and ROCK1 protein in lung tissues was detected by immunohistochemistry(IHC).The expression levels of Rho A and ROCK1 proteins in lung tissue were detected by Western blot.Morphological study of lung tissue specimens,pulmonary edema,pulmonary inflammatory cell infiltration,pulmonary hemorrhage were observed under HE staining.ResultsThe breathing and activity of rats in different doses of the heparin intervention group were significantly reduced compared with the CLP group,but local injection and nostril bleeding occurred in the HUFH group;the partial oxygen pressure Pa O2 and p H values ??of the CLP and UFH groups were lower than those of the Sham group,while MUFH Compared with the CLP group,Pa O2 was significantly increased in the HUFH group and the HUFH group(P <0.05);Pa O2 was increased in the LUFH group compared with the CLP group,but the difference was not significant,and there was no statistical significance;the difference in partial carbon dioxide pressure Pa CO2 between the groups There was no statistical significance(P> 0.05).Compared with theSham group,the levels of TNF-α,IL-1β,and IL-6 in the CLP group were significantly increased,and significantly higher than 4h at 24h(P <0.05);heparin The treatment group reduced the levels of inflammatory factors to different degrees.Compared with the CLP group,the levels of inflammatory factors in the HUFH and MUFH groups were significantly reduced,and the difference was statistically significant(P <0.05).Pathological observations of lung tissue found that The alveolar structure of the Sham group was clear and uniformly spaced,and there was no obvious infiltration and exudation of inflammatory cells.In the CLP group,part of the alveoli of the lung tissue atrophied or disappeared.In addition to the large number of inflammatory cells,interstitial congestion,edema,and alveolar wall thickening were also observed.In the CLP + heparin group,the lung tissue damage was reduced compared with the CLP group,the alveolar cavity was clear,the alveolar septum was slightly edema,and the inflammation was mild;the Sham group and the heparin intervention group had lower expression levels of Rho A and ROCK1 in lung tissue;The expression level was significantly higher than that in the Sham group.The heparin intervention group could inhibit the increase of Rho A and ROCK1 expression in lung tissue.Compared with the Sham group,the expression of Rho A and ROCK1 in the sepsis group was significantly upregulated,and the difference was significant(P <0.05).Compared with the CLP group,the expressions of Rho A and ROCK1 protein in the heparin intervention group were decreased(P <0.05).Conclusions1.The pathological lung tissue、pulmonary edema and inflammatory cells infiltrated was found 24 hours after CLP in rats.the expression levels of inflammatory factors in blood and lung tissues were increased,and syndrome combined with ALI modeling success.2.Heparin treatment group reduced the degree of ALI inflammation,reduced pulmonary edema caused by CLP and improved lung gas exchange function.3.Medium-dose UFH treatment group is better than high-and low-dose UFH treatment group.
