objective: To explore the protective effect and mechanism of Chai Huang Qing Yi Huo Xue Fang on liver injury of severe acute pancreatic in rats model by detection serum amylase(AMY),serum lipase(LPS),white blood cells(WBC),interleukin-1β,interleukin-6(IL-6),interleukin-18(IL-18),alanine aminotransferase(ALT),aspartate aminotransferase(AST),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),peroxidation Expression of catalase(CAT)of pancreas and liver tissue,malondialdehyde(MDA),and the expression of nuclear transcription factor-κB(NF-κB)and heme oxygenase-1(HO-1)in Severe acute pancreatitis rats.Methods : 128 male SPF SD rats,weighing 220g-280 g,were randomly divided into 4 groups:Blank(n = 32),Sham(n = 32),and SAP(n= 32),Chai Huang Qing Yi Huo Xue Fang group(chaihuangzu)(n = 32),Before the experiment,rats were fasted for 12 hours,retrograde injection of 5%(g / ml)sodium taurocholate solution(0.1ml / 100g)through the bile pancreatic duct to make severe acute pancreatitis rats model.Sham group and SAP group were gavaged with saline,Chai huang group were treated with Chai Huang Qing Yi Huo Xue Fang(1.2g/ kg),which was made into Liu Jin Gao,Rats fed with saline and Chai Huang Qing Yi Huo Xue Fang per 6 hours.Blood,pancreas and liver tissues were collected from 8 rats in each group at 6h,12 h,24h,and 48 h after modeling;Morphological observation and HE staining of pancreas and liver in each group of rats;Detecte serum AMY,LPS,ALT,AST,blood WBC and SOD,GSH-Px,CAT,MDA in fresh pancreas and liver tissues by biochemical assay.Detecte serum IL-1β,IL-6,IL-18 by enzyme-linked immunosorbent assay.Detect the expression of NF-κB,p-NF-κB and HO-1 in pancreas and liver tissues by Western Blot.Immunohistochemical staining was used to detect the expression of NF-κB in liver tissues;Real-time quantitative PCR was used to detect the expression of HO-1 m RNA and NF-κB m RNA in fresh liver tissues.Results: 1.Compared with the blank group,the sham operation group had no obvious changes in the pancreas and liver tissues observed by eyes and optical microscope,and there were no significant differences in serum AMY,LPS,ALT,AST,IL-1β,IL-6,IL-18 and blood WBC levels.There were no significant differences in the expression of SOD,GSH-Px,MDA,CAT,NF-κB,p-NF-κB,and HO-1 in pancreatic and liver tissues,and no significant differences in the expression of HO-1 m RNA and NF-κB m RNA in liver tissues;2.Compared with the sham group,pancreas and liver tissues of the Chaihuangzu and the SAP group had obvious pathological changes observed by eyes and optical microscope.serum AMY,LPS,ALT,AST,IL-1β,IL-6,IL-18 and WBC levels were significantly increased(p <0.05),expression of SOD,GSH-Px,CAT of pancreas and liver tissues decreased(p <0.05),while MDA level increased(p <0.05),expression of NF-κB,p-NF-κB,HO-1increased in pancreas and liver tissues(p <0.05),expression of HO-1m RNA,NF-κB m RNA increased in liver tissues(p <0.05);3.Compared with the model group,pancreas and liver tissues damage in the Chaihuangzu was improved,and serum AMY,LPS,ALT,AST,IL-1β,IL-6,IL-18,and WBC levels were reduced(p <0.05),expression of SOD,GSH-Px,and CAT increased in pancreas and liver tissues(p <0.05),while the MDA levels decreased(p<0.05).Expression of NF-κB,p-NF-κB decreased(p <0.05),and HO-1 expression increased in pancreas and liver tissues(p <0.05).Expression of NF-κB m RNA decreased(p<0.05),and expression of HO-1 m RNA increased in liver tissues(p<0.05).The differences are statistically significant.Conclusion: 1.Chai Huang Qing Yi Huo Xue Fang has protective effect on liver injury in SAP model rats;2.Chai Huang Qing Yi Huo Xue Fang may protect liver injury in SAP model rats by reducing the levels of pro-inflammatory inflammatory factors,down-regulating the expression of NF-κB and up-regulating HO-1.
