ObjectiveTo investigate the mechanism of Chinese Toona leaf extract on retinal injury induced by high fat diet in mice.Methods40 male C57BL/6J mice,randomly were divided into 4 groups:Normal group(NC group),high-fat diet feed group(HF group),and low-dose TSLE and high-dose TSLE group(L-TSLE group and H-TSLE group).Mice were examined by electroretinogram(ERG),optical coherence tomography(OCT),fluorescein fundus angiography(FFA);Blood lipids,HE staining,immunofluorescence,and western blot were used to check the expression of eptin receptor,jak2,stat3,vegf,socs3 protein in different treatment group.ResultsAt 30th week,The results of blood lipid tests showed that TSLE upregulated HDL-C and downregulated TG,TC,LDL-C;HE staining appeared that TSLE improve abnormal arrangement of retinal core and outer nucleus cells induced by high-fat diet feed,and recovered the structure of retinal pigment epithelium;FFA test confirmed that TSLE shortened the prolonged filling time of fluorescein induced by high-fat diet feed;The data of ERG test presented that the TSLE reduced high-fat diet feed induced-impairment of retinal function,resulted in increased a-wave and b-wave amplitude,which means recovering of retinal functions;in addition,Western blot results confirmed that TSLE downregulated expressions of lipid metabolism-related leptin receptors and upregulated JAK2,STAT3,VEGF,SOCS3 proteins.ConclisionTSLE played a therapeutic protective effect on retinal injury induced by high-fat diet feed in mice,which could be regulated through the jak/stat signal pathway. |
