Objective To explore the expression and clinical prognosis of MUC4 and MUC16 in liver cancer and its relationship with MUC1Methods 1.Ualcan database was used to analyze the expression of MUC1,MUC4,MUC6,MUC16 in liver cancer tissues online.2.Kaplan-Meier Plotter database was used to analyze and draw the survival curve,and Log-rank test was used to compare the survival prognosis of patients with high and low expression groups MUC1,MUC4,MUC6,MUC16.3.Online analysis of Meth HC database on the methylation of MUC1,MUC4,MUC6,and MUC16 in liver cancer tissues and normal liver tissues.4.GEPIA database online analysis of the relationship between MUC1,MUC4,MUC6,MUC16 expression levels in liver cancer tissues.5.Ualcan database further analyzed on-line the co-expression of MUC1,MUC4,MUC6,MUC16 in liver cancer tissues.6.c Bioportal database online analysis of MUC1,MUC4,MUC6,MUC16 gene mutations and co-occurrence of mutations in patients with liver cancer and their survival prognosis.7.STRING database finds proteins that interact with MUC1,MUC4,MUC6,MUC16.Results 1.The m RNA expression levels of MUC1 and MUC6 in liver cancer tissues are lower than those in normal liver tissues(both t-tests P <0.05),and the m RNA expression levels of MUC4 and MUC16 in liver cancer tissues are higher than those in normal liver tissues(both t-tests P<0.05).2.The high expression of MUC1 in liver cancer was negatively correlated with the overall survival(Overall Survial,OS)(P=0.011)and disease-specific survival(DSS)(P=0.0091)of patients with liver cancer;The high expression in liver cancer was negatively correlated with OS of patients with liver cancer(P=0.01);the high expression of MUC6 in liver cancer was negatively correlated with progression-free survival(PFS)in patients with liver cancer(P=0.038);The high expression of MUC16 in liver cancer was positively correlated with OS,DSS,and PFS in patients with liver cancer,and the P values were(P=3e-04),(P=9.2e-06),(P = 1.7e-06)).3.The methylation level of MUC1 in liver cancer tissues is higher than that in normal liver tissues,but the difference is not statistically significant;the methylation levels of MUC4,MUC6,and MUC16 in liver cancer tissues are significantly lower than those in normal liver tissues.Statistical significance(P<0.005).4.Analysis of the GEPIA database found that the expression level of MUC1 in liver cancer tissues was positively correlated with the expression levels of MUC4,MUC6,and MUC16,the expression level of MUC4 was positively correlated with the expression levels of MUC6,MUC16,and the expression level of MUC6 There was a negative correlation with MUC16 m RNA expression level.5.Collaborative analysis showed that MUC1,MUC4,MUC6,and MUC16 were significantly correlated with TC2 N,TSPAN1,CSGALNACT1,and STS,respectively.Most mutations in MUC1 are amplification mutations,and most mutations in MUC4,MUC6,and MUC16 are missense mutations.Mutations of the four genes MUC1,MUC4,MUC6,and MUC16 are related to survival prognosis(P=6.732E-3).At the same time,the four genes MUC1,MUC4,MUC6,and MUC16 have co-occurrence.6.Interacting proteins are MUC5 B,MUC5AC,MUC6,MUC7,MUC13,MUC15,GALNT4,MUC1,MUC21,GACNT6,B3GNT5,MUC12,MUC17,and MUC20.These proteins also actively participate in the regulatory network.Conclusion 1.MUC1 and MUC6 may inhibit the occurrence and development of liver cancer,MUC4 and MUC16 may promote the occurrence and development of liver cancer.2.MUC1,MUC4 and MUC16 are related to the overall survival of liver cancer patients,while MUC6 is not related to the overall survival of liver cancer patients.3.MUC1,MUC4,MUC6,and MUC16 gene mutations are involved in the occurrence and development of liver cancer,and patients with mutations have a worse prognosis.4.The methylation levels of MUC1,MUC4,MUC6,and MUC16 in liver cancer help to clarify a series of problems such as the occurrence,development and prognosis of liver cancer,and provide new ideas for early diagnosis and advanced gene therapy of liver cancer.5.Analysis of the correlations between MUC1,MUC4,MUC6,MUC16 and their interacting proteins and co-expression analysis will help provide new therapeutic targets for the treatment of liver cancer.
