Microphthalmia-associated transcription factor(Mitf)is an important transcription factor in melanocytes and an important factor in the synthesis of melanin.It controls the synthesis,transport and distribution of melanin by regulating the Tyrosinase(Tyr)gene family.By software prediction,it was found that there were sites on the 3’UTR of Mitf that could bind to miR-182-5p.There are also sites on circLuc7 L that bind to miR-182-5p.The relationship between circLuc7L/miR-182-5p/Mitf and its effect on B16F10 cells were verified by experiments.The expression sites of circLuc7 L were determined by in situ hybridization,and the biometric prediction results were verified by dual luciferase gene reporting technique and gene transfection technique.The effect of circLuc7L/miR-182-5p/Mitf regulatory axis on the production and migration of cytochrome in B16F10 cells was preliminarily studied.The results showed that circLuc7 L was specifically expressed in the cytoplasm of B16F10 cells,and there was a targeted binding relationship between circLuc7 L and miR-182-5p,and between Mitf and miR-182-5p.Moreover,miR-182-5p could inhibit the expression of Mitf and its downstream genes at the post-transcriptional level by binding to the3’UTR region of Mitf.circLuc7 L could indirectly regulate the expression of Mitf and its downstream genes by adsorbing miR-182-5p.circLuc7 L could significantly inhibit the migration of B16F10 cell line.
