Abjective: To investigate whether exercise-induced renal ischemia-reperfusion and renal dysfunction may occur in rats under high-intensity exhaustion exercise,and its relationship with apoptosis;In addition,resveratrol was used as a nutrition intervention to observe whether resveratrol had protective effects on renal dysfunction caused by exercise-induced renal ischemia-reperfusion and whether the protective effect was related to the anti-apoptotic activity of resveratrol.Methods: 32 SD rats were divided into a quiet control group(Con),a quiet +resveratrol group(Rsv),an exhausted exercise group(Ex),an exhausted exercise +resveratrol group(Ex + Res),There are four groups of eight in each.Exercise group and exercise + resveratrol group rats for three days of exercise preconditioning.In the formal experiment,the treadmill speed increased from 10 m / min to 35 m / min to maintain training until exhaustion,exercise 1 time / d,5d / week,a total of 4 weeks.The resveratrol group and the exercise + resveratrol group were administrated with resveratrol once per day,50 mg / kg,and the volume was 5 m L / kg.The quiet control group and exercise group were given an equal volume of 0.5% sodium carboxymethyl cellulose solution.After 24 hours of the last training,the rats were sacrificed,the blood was collected from the abdominal aorta,centrifuged at rest,and the upper serum was collected and aliquoted and stored in a-20 ℃ refrigerator for storage.Take the left and right kidneys and remove the surrounding fascia tissue,longitudinally cut the left kidney,and then immerse it in 4% paraformaldehyde fixative.Put the right kidney in pre-chilled saline and wash the blood,then Quickly put it into liquid nitrogen for temporary storage,and then freeze it for testing.Serum creatinine Scr was measured by alkaline picric acid method,and blood urea nitrogen BUN was measured by ultravioletglutamic acid dehydrogenase method.The protein expressions of P53,acetylated P53,and Caspase3 were detected by Westernblot.SIRT1 deacetylase activity was measured by double antibody sandwich enzyme-linked immunosorbent assay(ELISA).This study used SPSS 21.0 statistical software to process the data.One-way ANOVA was used for the analysis between groups.The data results were expressed as mean ±standard deviation(x ± s).P <0.05 indicates a significant difference,and P <0.01 indicates a significant difference.Result: In this experiment,the levels of SCr and BUN in the exercise group were significantly higher than those in the quiet control group(P <0.01).The SCr and BUN levels in the exercise + resveratrol group were significantly lower than those in the exercise group(P <0.01).The expression levels of Caspase3(P <0.05),P53(P<0.01),and acetylated P53(P <0.01)in the exercise group were significantly higher than those in the quiet control group.Caspase3(P <0.01),P53(P <0.05),acetylated P53(P <0.01),and acetylated P53 / P53 ratio(P <0.01)were significantly lower in the exercise + resveratrol group than in the exercise group.SIRT1 protein deacetylase activity test results were significantly higher in the resveratrol group than in the quiet control group(P <0.01).The exercise + resveratrol group was significantly higher than the exercise group(P <0.01).Conclusion:1 Up-regulation of P53 protein,acetylated P53 protein,and Caspase3 protein in rat kidney tissues caused by exercise-induced renal ischemia-reperfusion during high-intensity exercise is the cause of renal dysfunction.2 Resveratrol can reduce Caspase3 protein expression by down-regulating the expression of P53 protein and deacetylating the P53 protein by SIRT1 protein,thereby inhibiting renal dysfunction caused by exercise-induced renal ischemia-reperfusion in rats. |
