| ObjectivesIn this study,we investigated the effects of Insulin-like Growth Factor-1(IGF-1)and RBM8A on the proliferation and differentiation of retinal progenitor cells(RPC),which may provide a new way for the treatment of degenerative retinal diseases.Methods1.Under proliferation conditions,morphological observation,CCK8 analysis,qPCR,Western blot and immunocytochemistry were preformed to evaluate the effect of IGF-1 on the proliferation of RPCs;Under differentiated conditions,qPCR and immunocytochemistry and Western blot were preformed to investigate the potential fates of the IGF-1-treated RPCs.2.RPCs were transfected with Rbm8a plasmid or Rbm8a small interfering RNA(siRbm8a)and cultured under the condition of proliferation or differentiation,the cells were collected at the designated time points in the culture process.qPCR,Western blot and immunocytochemistry were used to detect the expression of genes and proteins related to proliferation and differentiation of RPC.3.Results1.Our data demonstrate that IGF-1 can activate PI3K/Akt and MAPK/ERK downstream signaling pathways through IGF-1R to promote the proliferation of RPC significantly;Besides,IGF-1 pretreated RPCs have stronger ability of differentiation towards retinal neuron under differentiated conditions.2.RBM8A decreased accordingly during RPC differentiation,which play an important role in the proliferation and differentiation of RPCs.And our data also showed that Rbm8a can enhance the proliferation of RPCs markedly.Meanwhile,under differentiated conditions,the Rbm8a-knockdowned RPCs were differentiated into more retinal neurons efficiently.Conclusion1.IGF-1 promoted the proliferation of RPC significantly through the activation of receptor IGF-1R and the PI3K/Akt and MAPK/Erk downstream signaling pathways.In addition,pre-treatment of IGF-1 can promote RPC differentiate into retinal neurons.Suggesting that IGF-1 play an important role in promoting the proliferation of RPC and improving RPC’s differentiation ability.2.In the process of proliferation and differentiation of RPC,RBM8A,as an important regulatory factor,promote the proliferation of RPC remarkably and induce the differentiation of RPC under differentiated conditions.It provides a new experimental basis for the treatment of retinal degenerative diseases. |
