Expression And Clinical Significance Of MicroRNA-106b In Plasma And Tumor Tissue Of Patients With Laryngeal Squamous Cell Carcinoma

Objective Recent research suggests that miRNA plays an important role in the occurrence and development of tumors,and the expression level of miRNA has potential significance for the diagnosis of cancers.In this study,the expression level of miR-106 b in the tissues and plasma of laryngeal cancer patients was detected to analyze the correlation between miR-106 b level and the clinical and pathological characteristics of patients and explore the value of miR-106 b expression level in the early diagnosis of laryngeal cancer.Methods A total of 50 cases of laryngeal cancer patients hospitalized in the department of otolaryngology,Zhongnan Hospital of Wuhan University from December 2016 to December 2018 were selected as the experimental group,while 50 cases of volunteers who underwent physical examination in our hospital were selected as the control group.Real-time quantitative PCR(q RT-PCR)method was used to detect the expression level of miR-106 b in tissue samples of laryngeal cancer patients and plasma samples of all subjects,the clinicopathological characteristics of patients with laryngeal cancer were collected.Results The relative expression level of miR-106 b in plasma samples from LSCC patients was(2.05±0.73),which was significantly higher than in those of the controls(1.36±0.50).The relative expression level in LSCC tissues was(4.27±1.37),which was more up-regulated than in the non-tumor tissues(2.49±0.80),the differences were statistically significant(P<0.05).The expression level of miR-106 b showed a highly linear positive correlation in the plasma and tissues of laryngeal cancer patients(r=0.712,P<0.05).The expression level of miR-106 b in tissues samples was obvious related to tumor differentiation,lymph node metastasis and TNM stage(P<0.05).The expression of miR-106 b in plasma samples has significant correlation with lymph node metastasis and TNM stage(P<0.05).In early-stage LSCC(stageⅠ)patients,the result of ROC curve showed that an area under the curve(AUC)of tissue miR-106 b was 0.756,with a sensitivity of 63.6% and a specificity of 81.8%.The AUC for plasma miR-106 b was 0.731,with a sensitivity of 63.6% and a specificity of 77.3%.Conclusions The expression of miR-106 b was up-regulated in plasm and tumor tissues of patients with laryngeal cancer.The expression level of plasma miR-106 b was significantly related to the TNM stage and lymph node metastasis,suggesting that the expression level of plasma miR-106 b might be a novel biomarker for the early diagnosis of laryngeal cancer.
