| Objective: To explore the changes of serum pNF-H and UCH-L1 protein levels in neonatal HIBD rats by establishing an animal model to simulate the occurrence and development of neonatal HIBD.Methods: 252 7-day-old neonatal SD rats were randomly divided into experimental group(HIBD group)and control group(sham-operated group and blank control group).The above-mentioned three groups were randomly divided into 7 sub-groups: 6h、12h、24h、48h、72h、5d and 7d groups,with 12 rats in each sub-group.HIBD models was prepared by the method of Reference in HIBD groups,that’s the left common carotid artery was ligated and hypoxic for 2 hours.In the sham operation group,the left common carotid artery was separated but not ligated,and hypoxic treatment was not performed after suturing the skin.While the blank control group with nothing(no hypoxic-ischemic treatment).Blood was collected at different time points in each group of newborn rats,and to detect serum concentrations of pNF-H and UCH-L1 by ELISA method.Results: Compared with the control group,the neonatal rats in HIBD groups showed Significant differences in motor behavior and neurological deficits in different degrees at 2 hours after the model was established.The serum levels of pNF-H and UCH-L1 protein in HIBD groups were obviously at a high level compared with those in control group at different time points(P < 0.01),and the concentrations of pNF-H and UCH-L1 protein in different subgroups of HIBD group were significantly exceed than those in control group(P < 0.01),while there was no significant difference in the serum levels of pNF-H and UCH-L1 protein between different subgroups of the control groups(P > 0.05).The serum levels of pNF-H protein in HIBD groups increased significantly at 6h and decreased gradually at 72 h,while UCH-L1 increased significantly at 6h and decreased gradually after 48 h,both of all decreased gradually after 7 days.Conclusions: The serum levels of pNF-H and UCH-L1 increased significantly in the early stage of HIBD,which could be used as a detection index in the early auxiliary diagnosis of HIE.It is helpful to guide the early targeted intervention and treatment in clinic,so as to reduce the mortality and disability rate. |
