Objective:The typical cases of lung cancer treated by Yiqi Sanjie Pill were summarized.The components of Yiqi Sanjie Pill were analyzed by LC-Q-TOF-MS,and the effective components of anti-cancer were screened.Method:1.By sorting out and analyzing the cases of lung cancer with partial remission(PR)and above treated by Professor Lin Lizhu’ s combination of traditional Chinese and Western medicine and the cases of middle and advanced lung cancer treated by maintenance of traditional Chinese medicine,this paper summarizes the experience of Professor Lin Lizhu in treating lung cancer with Yifei Sanjie Pill.2.Yiqi Sanjie Pill was pretreated,and the best anti-tumor active fraction was selected through cytotoxicity experiments.Components analysis of the active fractions was carried out.Combined with multidimensional statistical analysis and molecular docking,10 components were selected as potential anti-tumor components.Result:Professor Lin Lizhu believes that the main pathogenesis of lung cancer is deficiency of vital energy,phlegm and blood stasis."Deficiency,poison,blood stasis and nodule" are the pathological causes of lung cancer.Therefore,the treatment of lung cancer should be based on benefiting the lung,dispersing the nodules and resolving phlegm.For all stages treatment of lung cancer,as well as oral targeted therapy patients can use Yifei Sanjie pill as the main prescription,combined with the patient’s constitution and other traditional Chinese medicine treatment.And patients also can take Yifei Sanjie pills for long-term maintenance treatment.The active fractions of Yifei Sanjie Pill were pretreated and divided into five active fractions:total extract,30%,50%,70%and 100%methanol solid phase extraction.Cytotoxicity experiments were carried out on five samples.Among them,100%methanol solid phase extraction(YFSJP-M4)had the best anti-tumor activity.Principal component analysis was used to analyze the peak area and retention time of four active fractions and total extract,combined with YFSJP-M4.The YFSJP-M4 was analyzed by liquid chromatography and mass spectrometry.The top 10 possible anti-tumor active ingredients were 4-hydroxy-3,5-dimethoxybenzoic acid,3-[(4-O-D-glucosyl)-phenyl]-2-acrylic acid,Ganoderenic acid D,myristic acid,rosmarinic acid,gallic acid,benedine,Ganoderma acid C2,α-Linolenic acid and isorhamnetin.Based on our previous studies,GRP78 protein is considered to be a key protein in regulating endoplasmic reticulum stress-induced apoptosis of cancer cells.Therefore,GRP78 protein is applied to molecular docking.The results show that isorhamnetin,berberine and rosmarinic acid are possible anti-tumor components.CONCLUSION:Yifei Sanjie Pill is Professor Lin Lizhu’s experience in treating lung cancer based on years of experience,which has a wide range of clinical applications.It can significantly improve symptoms and quality of life for lung cancer patients at all stages.To a certain extent,it can prolong the progression-free survival period and the survival period of palliative treatment for advanced patients.Through the extraction and screening of the active fraction of Yifei Sanjie Pill,it was concluded that the YFSJP-M4 had the best anti-tumor effect.The compounds were identified by bioactive fractions screening combined with LC-MS and predicted by molecular docking simulation.Isorhamnetin,Friedrine and Rosmarinic acid were identified as effective antineoplastic substances.
