ObjectiveTo investigate the expression of BTB/POZ domain containing protein 7 in papillary thyroid carcinoma and its correlation with clinicopathological features.The effects of BTBD7 on PTC invasion and metastasis and its possible mechanisms were further explored.Provide new reference indicators for PTC gene diagnosis,prognosis assessment and provide therapeutic targets for treatment.ObjectiveWe selected 145 cases of thyroid papillary carcinoma and adjacent cancer specimens which were diagnosed by pathological diagnosis.Then tissue chips were made and expression of BTBD7 protein were detected through immunohistoch-emistry methods.Based on that positive expression depends on the appearance of obvious brown-yellow particles in the cytoplasm,BDBT7 is mainly expressed in cytoplasm.Statistical analysis software SPSS 23.0 was used to analyze the relationship between BTBD7 and the pathological features of papillary thyroid carcinoma.In order to confirm the effect of BTBD7 on cell invasion and migration,this study performed overexpression and interference of BTBD7 in two PTC cell lines,and detected changes in BTBD7 at protein and m RNA levels,and expression of EMT-related proteins(E-cadherin,N-cadherin,Snail2).Transwell assay was used to detect invasion and migration of two PTC cell lines.Results1.The expression of BTBD7 in papillary thyroid carcinoma was significantly higher than that in adjacent tissues(p<0.05).2.The expression of BTBD7 was not significant associated with age,gender and tumor size,but was related to tumor invasion,lymph node metastasis,distant metastasis and TNM stage(p<0.05).3.The number of invasion and migration of cells of si BTBD7 was significantly lower than that of the si NC group;compared with the control group,over-expressedBTBD7 cells was significantly increased in the number of invasion and migration.While BTBD7 was up-regulated,the expression of N-cadherin and Snail2 was down-regulated.And after over-expression of BTBD7,the expression of E-cadherin was down-regulated and the expression of N-cadherin and Snail2 was up-regulated.Conclusion1.The expression of BTBD7 in papillary thyroid carcinoma is significantly higher than that in adjacent tissues.And associated with tumor invasion,lymph node metastasis,distant metastasis,and TNM staging.2.BTBD7 induces EMT in papillary thyroid carcinoma,which promotes invasion and migration of papillary thyroid cancer.
