Study On The Quality Standard Of Salt Morinda Officinalis Based On The Fingerprints-pharmacology Relationship Of Reinforcing Kidney-yang

Morinda officinalis How.Radix euphorbiae,one of the four southern herbs in China,has fleshy roots.Euphorbia officinalis has the effects of nourishing kidney and Yang,strengthening muscles and bones and removing rheumatism.It can be used in clinical treatment of impotence and spermatorrhea,infertility due to cold in the uterus,irregular menstruation,cold pain in the abdomen,rheumatism and numbness,weakness and weakness.Morinda officinalis is a kind of traditional Chinese medicine with dual use as medicine and food.At present,the development of treatment and health care products with morinda officinalis as raw material has become a focus of people’s attention,especially the research and development of products with the function of tonifying kidney and Yang.Clinically,morinda officinalis is often used as the preparation for tonifying kidney Yang,and the cooperative enterprise in this project has developed a new product with the effect of tonifying kidney Yang by taking morinda officinalis as the main medicinal flavor.The 2015 edition of Chinese pharmacopoeia used the content of nestose to evaluate the quality of halberd officinalis,and the quality of the prepared slices of halberd officinalis used for tonifying kidney Yang could not be fully controlled.At present,the effective ingredients of halberd for tonifying kidney Yang are not completely clear.Therefore,to discuss the effective ingredients of halberd for tonifying kidney Yang and then guide the formulation of internal quality control standards of halberd for enterprises with effective ingredients of tonifying kidney Yang will provide strong support for the quality control of enterprise products.Department of traditional Chinese medicine(TCM)spectral efficiency will indicate the active ingredient of characteristic peak of traditional Chinese medicine fingerprint with the corresponding efficacy results,the efficacy results associated with the change of chemical composition,the chemical content of Chinese traditional medicine and the relationship between efficacy,thus effectively control quality of traditional Chinese medicine and compound preparations,explore new methods of traditional Chinese medicine medicinal material base.Based on the spectrum-effect research strategy,this paper carried out the correlation study between the chemical components of halberd and the effect of tonifying kidney-yang,clarified the effective components of tonifying kidney-yang,and then guided the establishment of the quality standard of halberd,and provided reference basis for the further establishment of the quality standard of medicinal materials and decoction pieces of halberd.This paper mainly completed the following research work:1.according to a study in the early stage of the laboratory fingerprint and total sugar,total iridoid,total anthraquinone content determination method,the acquisition of the 23 after production batches of different origin herbs salt fingerprint detection and determination of composition of three types of content,according to the result of the experiment selected 10 batches of larger difference of salt ji days,at the same time,the early stage of the reference laboratory to optimize the extraction process preparation extract,and detect the extracts from 10 batches of UPLC-DAD fingerprint,gain mutual fengfeng area,as the late spectral analysis data.2.using adenine to kidney Yang deficiency model mice,thyroidstimulating hormone(TSH)in mice and free thyroxine(FT4),free iodine thyroid original glycine(FT3),testosterone(T),cortisol(;CORT),luteinizing hormone(LH),follicle-stimulating hormone(FSH)as evaluation index,10 batches of different origin salt ji days pharmacodynamics study of kidney Yang.TSH,f T4,f T3,;CORT,LH,T,the results showed that compared with model group,10 batches of salt ji days extracts were significantly or extremely significantly reduce FSH levels(P < 0.05 or P < 0.01),can significantly or extremely significantly improve TSH,f T4,f T3,T,;CORT,LH levels(P <0.05 or P< 0.01),suggesting that the kidney Yang deficiency syndrome in mice were improved,but the efficacy of effect size differences.3.Analytic hierarchy process(ahp)was used to determine the weight coefficient of each pharmacodynamic index,and grey relational degree analysis and partial least squares regression analysis were conducted on each chemical component represented by each characteristic peak,each pharmacodynamic index and the total pharmacodynamic effect in the fingerprint.Results show that the grey correlation degree analysis of UPLC-DAD fingerprint 24 of the 25 characteristic peak peak correlation of total efficacy index above 0.6,three peak correlation of total efficacy index above0.8,shows that most of the fingerprint related to chemical composition and the kidney Yang efficacy,but the composition degree of contribution to the total efficacy,the top five correlation peak is 11 >17>13>18> 20,8、11、17 peak and peak correlation is greater than 0.8,13 for unknown chemical composition,has a large research value;No.8 peak and no.20 peak are respectively crystalline lanoside and chedelic oxalic acid,both of which are iridoids.In the partial least squares regression analysis,there were 12 characteristic peaks of25 characteristic peaks of uplc-dad fingerprint that were positively correlated with the total drug efficacy,and the top 5 characteristic peaks of positive correlation were 20>21>17>1>19,among which the chemical components represented by characteristic peaks no.20 and no.21 were all iridoids.The results of the two methods showed that iridoid plays an important role in the improvement of kidney-yang deficiency,suggesting that iridoid should be added as quality control index when improving quality standard of halberd.4.14 batches of halberd from different places were selected for quality standard study according to the results of preliminary laboratory study and spectral effect study in this paper.In accordance with the quality standards of halberd officinalis in China pharmacopoeia 2015,14 batches of halberd officinalis from different places were tested for traits,TLC identification,water content,total ash content,water soluble extract content and nessose content.The contents of total sugar,total iridoid and total anthraquinone were determined.UPLC was used to detect the contents of four kinds of iridoid and the fingerprint of halberd salt was established.Through comprehensive analysis,a relatively comprehensive and reliable internal purchase standard of halberd salt has been established,which provides a guarantee for strict control of product quality.To sum up,this paper USES the results of the basic research on tonifying kidney and strengthening Yang of halberd officinalis for guiding the quality standard research on halberd officinalis,and plays a demonstration role in the new research model of quality standard system of traditional Chinese medicine.The results of this study will have important guiding significance for further improving the quality standard system of morinda officinalis and other decoction pieces.
